Yes that will keep it from twisting. That is the way I still fill my spools of 
Copper Wire, Lead wire and the way I fill my spools for the Norlander Bobbins 
when I was commercial tying.
I bought all wire in bulk.

I've used mono tying thread and regular 6/0 uni thread with good results and no 
thread breakage.

What's the purpose of using regular mono? Economy?

Also, I never thought about winding material from one spool to the other in the 
same direction, does that practically eliminate twisting?
