I don't think you will notice a huge difference whether you have it set to dark mode or light mode. If you have a phone with OLED screen there might be a very small difference since apparently dark pixels use up less power, but it would be quite minimal. Of course you may as well set it to dark mode.
As for screen brightness I would recommend you set it to auto and then if you want to make the most of it you can turn on screen curtain by tapping 3 times with 3 fingers. As soon as iOS detects that screen curtain is on brightness will be automatically set to 0%, but if you ever need help with the phone then as soon as you turn off screen curtain with a 3-finger triple tap the screen brightness will be at a level which should make it easy for a sighted person to see it. You can of course also turn off Voiceover which will then automatically remove the screen curtain since that is a Voiceover feature so if Voiceover is not running screen curtain is also not there to black out the screen.
