First of all, you can manually backup to your computer even when iCloud backup is on, I do it all the time. As for your problem, does the computer recognize your iPhone? That would be the first thing to find out, do you get the USB device connect sound? If so, do you see your iPhone in "Computer" or "My Computer" if you are still on XP? If not, does your cable charge the phone when it's plugged into the wall charger? There is always a possibility that your cable is defective and it's usually the first thing you want to rule out. In iTunes itself, everything under "Devices" in the file menu is disabled, I will go back to Jonathan's excellent podcasts about iTunes 11 but I really don't know how to problem solve this one.
