Are you plugging your phone into a USB hub or a USB port directly on the computer? Anyhow, just try a different USB port and see if the computer charges the phone or recognizes it.

As for your problem, does the computer recognize your iPhone? That would be the first thing to find out, do you get the USB device connect sound? If so, do you see your iPhone in "Computer" or "My Computer" if you are still on XP? If not, does your cable charge the phone when it's plugged into the wall charger? There is always a possibility that your cable is defective and it's usually the first thing you want to rule out. I turned off the iCloud back up because I know I cannot back up on the computer if iCloud backup is enabled. When I go into "itunes WIFI Sync" in Settings I get the message that says, "to enable WIFI syncing with iTunes, connect to iTunes on your computer using a cable and click sync with this iPhone over WIFI." Just beyond that is a "Sync now" button; it's grayed out. Just past that it says, "Sync or review when Hips available." This is not a button. In iTunes itself, everything under "Devices" in the file menu is disabled, I will go back to Jonathan's excellent podcasts about iTunes 11 but I really don't know how to problem solve this one.
