Assistive touch will not interfere with voice over. Once you turn it on, a
small translucent button appears on the upper left corner of the screen. As
long as you do not touch this button your iPhone will perform as usual. When
you double tap the Assistive touch menu button, it will bring up a variety
of options. You will find the volume, lock, and mute controls under the
device button.

With Assistive touch you can control the iPhone on the touch screen
without using the actual hardware buttons like volume up and down, the
home button, mute button and of course the lock screen button. I plan to
use the Assistive touch feature in the future to reduce wear and tear on
the physical hardware.
BTW, it cost me seventy five dollars to get the lock screen button
repaired.
