There are three "Cache" folders in OSX.

Macintosh HD/System/Library/Caches
Macintosh HD/Users/*user*/Library/Caches
and
Macintosh HD/Library/Caches.

Move the contents of any of these folders to the trash (no need to empty trash however). All files and folders within the cache folders are safe *enough* to remove.

I know you may loose preferences such as any specifically changed default 'open with' settings for some documents and the computer will start up slowly after the first restart as the OS rebuilds these cache files.

Any particular ones or all of 'em?
Try deleting the contents of the "Cache" folders - usually works for me (if you can access them). You will most likely need to boot from another device though... Easy enough if you have another Mac and a FireWire cable!

Emac G4 1ghz running Leopard 10.5
Bought this computer about a month ago. Computer is in excellent nick and has been running perfectly. My wife installed a system update at the weekend (Auto Update) Now the finder won't load (Endless spinning wheel),No folders or disc images onscreen-only desktop picture. Have tried repairing permissions (Can only access disc utility via spotlight) The dock shows but does not activate anything except Dashboard. Spotlight indicates that all wife's folders and files are there. Zapped the PRAM - tried repairing permissions via disc utility on my Tiger install disc all to no avail. Am contemplating archiving Leopard and installing Tiger instead but am concerned about losing wifes folders and files.
CAN ANYONE HELP?
