Yes - boot from a known - safe OS to check for malware.
You could have something that is starting before the firewall, and AV
software and managing to hide from it
And most malware, having got onto one PC on a LAN will multiply/migrate to
all
Don't forget to disconnect all Client, and Server PC's from the LAN before
disinfecting/cleaning the PC's
and only connect each one back when you are sure it is clean.

Check the OS SP and fix levels as well as checking with the boot CD/DVD

However failure of a LAN connection may effect all PC's attached to that
LAN,

and could account for the slow start-up

Have you tried the PC systems when the LAN cables are detached from the PC's
