Dear all,

I am trying to model a weak dimer using intermolecular PCS. I have
intermolecular PCS from natural abundance protein bound to lanthanides
mixed with [U-15N] labeled protein that’s diamagnetic. Therefore, the PCS
that we are observing only come from the dimer.

Since the complex is weak, the PCS that I am measuring are scaled by the
small fractional population of the dimer and are smaller than they would be
for a 100% bound complex.

I am wondering if anyone has any experience with the appropriate way to
deal with the measured PCS to account for the small population of dimer?

I have an estimate of the Kd that I can use to calculate the fractional
population of dimer but I want to make sure I use the appropriate treatment
of the PCS.

Thank you in advance for any help.

Best regards,
Taylor
