If I understand your explanation right, the 550 error came from smtp.isp.com (which I assume is your ISP's smtp server). It can be done with one MTA, but the MTA has to be able to create multiple SMTP instances. For example, I use Exchange 2003. If I wanted to use it as an MTA and SMTP server, I would create what it calls a SMTP Virtual Server (you can have more than one of these). As long as it's on a different port from another one, it can exist. Anyways, I don't know if Groupwise has this capability, but it might and very roughly that's how you would do it.
