Hi I use Spotify Premium and it sounds like 320 kps MP3/4s to me in quality. Flacs, depending on compression level - which can still be variable using something like DBpoweramp - will still run at probably 700kps-1100kps, and therein lies your audiable difference. It will not be exponentially double or more the quality, but it still will be far less compressed than 320kps Spotify.
