The Kantronics generates data as audio to modulate the K3, and decodes the received audio to put characters on your screen. You will need audio in and out to/from the K3. The RS232 port in the K3 allows a computer to control the K3, and to get information from the K3 about frequency and operating mode. It does NOT include audio.
That's all I know about this box, so you'll have to study the manual to understand what to do with the serial port -- that is, does it go to the computer or to the radio, and how the Kantronics uses it.
