Information Store is all you need, and I recommed a nightly full backup. However, for your backup strategy, what is most important is the size of the information store, not the number of mailboxes, as the size of the information store and logs will dictate how much tape you need and whether you need to change it.
Implement deleted item recovery and deleted mailbox recovery for users to get their own deleted stuff back instead of backing up mailboxes. If you run a backup of the Information Store, Exchange runs. In fact, it must run for the backup to work. When backing up any files on the server, exclude the M: drive, \exchsrvr\*data directories, and the SMTP virtual server directories.
