I'd use a PS script and the EWS managed API. If you set it up right, you might be able to use a single transport rule doing a pattern match on the To: address, and copy them all to one mailbox. The script could read the messages, send a customized response based on the recipient address, and then hard delete the emails. Set it up as a scheduled task, and run it every half hour or so.

Have the rule delete the message but someone would have to log into that mailbox and purge those deleted messages. Thoughts on other ways to accomplish this?
