I often section brain sections at 50 microns. However, I use a sliding 
microtome. For me, those are the best at sectioning thick brain sections. 
Vibratomes are great too. I would use the cryostat as a last resort. I also 
agree that if it is too cold, it wont section correctly and will begin to 
crack. Monitor the tem,p and make sure that it is not too cold.

I need to cut 80 micron sections of frozen brain.
Tried today at -18 degrees C but the brains kept cracking. I read that the temp 
should be -3 to -13 degrees C. Has anybody cut sections this thick of brain? 
Can you pass along any words of inspiration?
