No cold start required as long as no volume containing SPOL allocations was moved to a different slot.
In my SHARE session "z/VM Installation - It's Installed, NOW What?" I stongly suggest that all SPOOL volumes be placed in the first few slots, the next slots be RESERVED (maybe a total of 10?) For SPOOL (slots are cheap), and comments provide clear reminders not to change the slot numbers. I suggest that SPOOL volumes be named so that there is no doubt that they contain SPOL allocations (e.g. VMSP01, VMSP02, etc), and at the top so that they don't' get sorted to a lower place in the future, and the slot numbers changed in a fit of housecleaning.
