The entire class C range that s2 would represent is in the "relay for addresses" 
section.

I have an MX record for the "public" IP address that is being relayed for.  For 
instance:
     The public IP of maildomain1.com is 216.160.208.102.  In the hosts file I have 
the address for maildomain1.com set up as 192.168.3.13.  This is per the instructions 
for setting up iMail as an SMTP Gateway instructions in the knowledge base.  The 
machine has two NICs.  One on the public network and one on the private network.  
maildomain2.com is 216.160.208.250 in the same class C block.  It is also on the same 
relay machine, except that the relay is actually hosting for this mail domain.

The bounce message I get is "maildomain1.com" not found, or something similar.

does the internal server have MX like this:

maildomain1.com    MX  10 Imail.server.com

who is "it"?  antecedent unknown, comment rejected

Can we see some imail logs when you send mail to the internal server?

you should have in NT hosts file:

ip.ad.re.ss     imail.domain.com    ; default ip address for
                                     ; default/nonvirtual Imail domain

ip.ad.rs.s2     maildomain1.com     ;

You also should have ip.ad.rs.s2 in smtp security, "relay for addresses", ime.

That otta work.
