I use the Linux version of WordNet under Cygwin for simple lookups. For complicated ones I use dictionary.com, again under Cygwin. But lately I'm increasingly impressed with Wiktionary, available through Research It.

I also have a list of nearly 400,000 words extracted from the SCOWL (Spell Checker Oriented Word List) package. This is just a list of words, but it's useful if you just want to verify the spelling of something.
