Tim,
May I suggest that the locking pin have a tapered blunt point on its business end.  That will absolutely ensure consistent registration as well as provide "lifetime" wear compensation (as the index hole "gets larger" due to wear, the taper still centers itself in the index hole.).  I am very interested in the indexing plate.  If you wanted to cover as many possibilities as possible with the fewest plates, two should do it:  one with 100 index holes and the other with 144 (2.5 degrees per hole)  I realize that these two plates are not the most user friendly way to go though.  That would entail more plates: 24, 36, 48, 60, 72, 144, 50, and 100.  OK, if you have a 144 hole plate, why a 72?  Because I will have to count half the number of index holes if using the plate to divide circle into 36 pieces.

Changing out the plate is just four screws that use the 3/16 wrench that comes with the machines.  If you lose your mind and crank, the plastic bevel gears will likely shred.  The holding power is pretty damn solid.  Backlash is not an issue since the lock and index plate is self aligning and accommodates wear.  You've hit most of my concerns with selling this tool.  
 
One vote for front mounted latch.

A concern I have is complacency.  There will be a lot of force able to be applied to the lock pin if someone just loses their mind and just starts to crank. 
What about back lash?  I guess the TK lockey lever will have to be part of the kit. 
How easy will it be to change out index plates?

One bit of input I would like from those interested in a locking system is which side do you want the lock handle to exit from.  My first choice is the side opposite the crank handle, but I'm also thinking the front would be good as well.
