Hi Jan, Do you have a possibility to check if you get dimerization at all and if the subunits are correctly folded? Maybe also some posttranslational processing is missing (signal sequences, *.ylation etc etc - any motifs present?). Have you considered to treat your proteins obtained from the Qiagen kit with eg sf9-cell free extract?
Is there a need for the cell free system or would the expression of tagged versions of your proteins in your favorite cells, followed by affinity purification, be an alternative?
