You don't say what version of software you are using. If it is Protel99SE, open the PCB file, go to File>CAM Manager, follow the wizard and select BOM as the output, you can then press F9 to generate the BOM or you can go to Tools>Generate CAM Files. Your BOM will be generated for all the components on the board.

If you are working with DXP2004, open the PCBDOC, go to Reports>Bill of Materials.

There are various add-ins, scripts, etc to tailor the BOM. The above is just the barebones way of getting the basic report.
