If the chroot jail is via the "chroot" column in master.cf, then this
is not the case, Postfix daemons enter the (Postfix queue) chroot jail
*after* loading main.cf. If "postfix start" starts from an external
(to Postfix) chroot jail, then indeed the main.cf file is found in
the external chroot jail.
