for so many users, I would use the vmailmgr package of Bruce G. :
http://www.vmailmgr.org/
You can then easily create a script which call the vaddusers
command (http://www.vmailmgr.org/docs/vaddusers.html).
Otherwise, there are other solutions : look at www.qmail.org.
