A simple desaturation is not very likely to be it. It used to be the case that the best B/W conversions were acheived with Rawstudio by adjusting the white balance, but nowadays it's easier to use the channel mixer as you say. (Note to developers: maybe consider adding a checkbox to keep pixel values constant and simply let the RGB sliders determine the ratio of each channel like in GIMP's built-in channel mixer.) What really matters is how happy you are with the results you get.
