 From my experience with Nikon cameras, I'd say the latter, but it could 
conceivably differ from camera to camera.

To answer that question you first have to define what is the correct B/W 
result. A simple desaturation is not very likely to be it. It used to be 
the case that the best B/W conversions were acheived with Rawstudio by 
adjusting the white balance, but nowadays it's easier to use the channel 
mixer as you say. (Note to developers: maybe consider adding a checkbox 
to keep pixel values constant and simply let the RGB sliders determine 
the ratio of each channel like in GIMP's built-in channel mixer.) What 
really matters is how happy you are with the results you get.
