Structural Biology of Membrane Proteins in the Laboratory of Cell Biology,

"Mechanistic investigation of cellular respiratory chain in drug resistance ".  
The project involves structural study of mitochondrial proteins that are 
targets of antimicrobial compounds.  It is anticipated that multiple structural 
techniques will be employed, involving the use of X-ray crystallography and 
Cryo-EM to illuminate structural aspects of target protein response to drug 
treatment.  Our group has assured access to a synchrotron beam line at APS and 
to an intramural EM facility equipped with a Krios microscope and Gatan K3 
detector.  While the candidate should have structural biology expertise, they 
are not expected to be proficient in all structural biology techniques. The 
successful candidate should have practical experiences in molecular biology and 
protein purification for target proteins production. Experience in

eBIC, a UK National Centre for cryo-electron microscopy, the partnership

This position is an opportunity to apply your cryo-EM knowledge to an array of

The Role:

The Associate Director, Structural Biology, will provide leadership and

  *   Collaborate with colleagues in lead discovery, biology, and chemistry to 
develop a structural biology strategy.

drug targets through working with external collaborators.

Requirements and Qualifications:

Education:

  *   PhD degree structural biology or biophysics

Required Experience:

  *   7+ years of drug discovery experience in a pharmaceutical/biotechnology 
setting;
  *   Demonstrated ability to solve difficult problems in protein 
crystallography, structural biology and biophysics
  *   Demonstrated competency in protein construct design and expression and 
purification strategies using bacterial, insect, and mammalian hosts.

Knowledge, Skills & Abilities:

  *   Strong knowledge of the drug discovery process, with deep experience in 
implementing hit identification strategies through candidate selection;
  *   Extensive knowledge of protein structure and function, with the ability 
to articulately communicate structural information to a broad audience.
  *   Experience with other structural techniques (e.g. electron microscopy, 
small-angle X-ray scattering) is preferred.

opportunity available in the Banfield Laboratory at the John Innes Centre in 
Norwich, UK.


We primarily study protein structure/function relationships important in the 
co-evolutionary arms race between plant pathogens and their hosts, with a 
particular emphasis on oomycetes such as the Irish potato famine pathogen 
Phytophthora infestans. This parasite continues to be a very serious problem 
for modern agriculture. Ultimately, in addition to making exciting fundamental 
discoveries, we expect our studies to drive development of novel methods for 
disease control.


The current opportunity would suit a post-doctoral level researcher interested 
in the molecular mechanisms that underpin microbial pathogenesis. The project 
will involve determining the structures of pathogen and host proteins alongside 
in vitro biochemical/biophysical interrogation of interactions, 
discovery/validation of protein activities and in planta studies (training 
available).

Background:

The Structural Genomics Consortium (SGC) is an international 
UK-Canadian-Swedish collaboration whose primary objective is to determine the 
3-dimensional structures of human and malaria proteins and release them into 
the public domain. The SGC is funded by the Wellcome Trust, a group of Canadian 
and Swedish funding agencies and GlaxoSmithKline. Since its establishment in 
2003 (the Swedish node added in 2004) it has built the infrastructure and 
technology platforms to determine the structures of ~200 proteins of medical 
relevance per year. The current Phase of funding for SGC Oxford ends in June 
2008. The application for renewal of the grant until June 2011 is currently 
under review. 

The SGC is one of the largest high throughput structural biology projects in 
the world, operating from three sites, one at the University of Toronto, one at 
the Karolinska University in Stockholm and one at the University of Oxford 
based at the Botnar Research Centre, which is located at the Nuffield 
Orthopaedic Centre in Headington, Oxford. All three sites have independent yet 
coordinated activities reflecting the scientific expertise and environment at 
each site. The Oxford site currently targets oxidoreductases, metabolic 
enzymes, kinases, phosphatases, G proteins and signalling components that are 
involved in human disease. A growing emphasis lies on transmembrane proteins.

The SGC Chief Executive, Dr Aled Edwards, has overall responsibility for 
implementation of the activity and is required to report quarterly to the Board 
of Directors (via the Chairman). Board members determine the strategic 
direction, milestones and administer the budget. The Chief Scientists, Dr 
Johann Weigelt, Dr Michael Sundstrom and Dr Cheryl Arrowsmith are coordinating 
the Stockholm, Oxford and Toronto operations, respectively. The Oxford site
