It was here for more than 7 days, then processed as usual into wax. IHC for Her2 has value as 2+. We fixed our BM biopsies at least 24 hours, but no more than 72 h in 10% NBF. Wash with tap water 30 mins. Decal into 14% EDTA (2Na salt) buffered with NaOH up to 7.0 overnight or weekend and test endpoint by weight loss weight gain method (special thanks to Gayle Callis!). Wash in tap water 1 hour. Process with isopropanols (IPA), mixtures IPA and mineral oil), paraffin wax. Microtomy at 3 microns. From sugh expensive blades can be done more very good sections (at 3 microns) from any difficult type tissue. These investments will pay off very quickly. Sandra, high profile blades was better for me, because they are harder and more durable than low profile of any manufacturer. Also I recommend for you to try Surgipath DB80HS. They must better than 818. I regret that not having bought a holder for a high profile blades. Leica makes the blade holders only for one type blades (low or high).
