I generally use CL Sturkey Extremus Low Profile Blades for pretty much 
everything...when I am having trouble with intestine sections, I use the 
DuraEdge Encore blade and it usually works very well.

[Histonet] Processing Protocol Using Mineral Oil

We have been using this protocol on mostly mouse tissues with great results!  
We run this on a VIP 3000.

70% Ethanol, room temp, 1 hour
100% Isopropyl Alcohol, room temp, 1 hour
100% Isopropyl Alcohol, room temp, 1 hour
100% Isopropyl Alcohol, room temp, 1 hour
5:1 Iso:Mineral Oil, 50 degrees, 1 hour
2:1 Iso:Mineral Oil, 50 degrees, 1 hour
100% Mineral Oil, 50 degrees, 1 hour
100% Mineral Oil, 50 degrees, 1 hour
100% Mineral Oil, 50 degrees, 6 hours
Paraffin, 60 degrees, 1 hour
Paraffin, 60 degrees, 1 hour
Paraffin, 60 degrees, 1 hour

Followed by a cleaning cycle after cassettes are removed and put into molten 
paraffin at the embedding center

I am purchasing my second Leica RM2255 automated microtome. Rene' is correct in 
saying that the carpal tunnel syndrome will eventually affect every advanced 
tech by using any microtome manually. On the RM2255, the flywheel additionally 
is automated. You can choose to use the flywheel manually or automated by the 
simultaneous pushing of two buttons. Nice feature for us oldies to ease us 
into automation or if you have a tiny specimen that you need to take extra care 
with and want to use manually.
Companies are very happy to send in a demo to try for a couple weeks.

The advantage of the so called automated microtomes (the only thing automated 
about them is the block advance) is that they alleviate wrist effort and in 
some ways prevent carpal tunnel syndrome that affects some histotechs (mostly 
of the senior persuasion).
I would go with the Leica.

Just curious if anyone has any experience with the Airfiltronix portable fume 
extractor filter?

I saw it in the Ted Pella catalog, item # 3120,  
http://www.tedpella.com/safety_html/3120.htm It is designed to be used with a 
microwave, but can be used anywhere, according to the catalog.  We are thinking 
about using something like this in our processing lab due to space constraints.

Any input is greatly appreciated.

[Histonet] Sakura VIP 3000 Tissue Processor

[Histonet] Murine Intestine Protocols

If anyone could share their protocols for intestine processing, embedding, 
cutting, etc I would greatly appreciate any input.  I am having trouble getting 
consistently good quality intestine sections and am always looking to improve.

[Histonet] Exam Prep Info/Guidance/Suggestions

I am about to begin preparing for the HTL certification exam.  (I supposse it 
is only fair to mention that this is will be my second attempt.  My first exam 
taken last month with a score of 382 out of a minimum passing score of 400).

As a research Histology Technician, a good majority of the material covered 
on the exam was new to me.  My prep for the first exam was to read 
Histotechnology, A Self-Instructional Text, 3rd Edtion by Carson and Hladik 
and then answer a lot of practice questions from the Board of Registry Second 
Edition Practice Questions book.  I focused mostly on the special staining info 
and images because of the high concentration of questions on the exam as well 
as my limited professionnal exposure to these...unfortunately, it was still the 
area i did most poorly in on the exam.

In order to go a little more in depth the second time around, my plan is to is 
to re-read each chapter ( of Carson), then assess my knowledge by being sure I 
can do each of the Chapter Objectives and then answer the questions in the 
self-assessement workbook, then answer the questions on that particular chapter 
topic from the Seond Edition of the Board of Registry Study Guide, Practice 
Questions for the Histotechnology Exams (The Purple Book).  I have other 
textbooks at my disposal, but as a mostly visual learner, I find Carson's 
approach to best get the information into my brain!  Again, once I feel 
comfortable with all the basic knowledge, I would focus mostly on staining.

Specifically, I would like feedback from anyone who has recently passed the 
exam about my approach, and suggestions for other ideas.  Also, I have very 
limited CAP/Joint Comission type knowlege and wonder if anyone can offer a 
resource for basics for someone like me in research who is not presented with 
the info and regulations on a daily basis!  Also, any suggestions for images 
online of special stains would be a great resource for me.

[Histonet] Zeiss Microm Rotary Microtome HM 325

[Histonet] Bar Coding and Sample Tracking

Looking for advice/comments/suggestions regarding sample tracking and data 
management.  I work for a small research institution doing mostly mouse/monkey 
tissues.  Up until now, we have been tracking samples with an excel 
spreadsheet, but increasing workload is making this more and more difficult.  
We average probably less than 500 blocks per month (very rough estimate).

Basically what I think we need is bar coding for samples and a centralized data 
storage place so that all the information available for a single sample is in 
the same place, and can be accessed by anyone anywhere, but not altered (for 
GLP compliance) as well as capability to expand as our workload varies.

This is my first step in looking into this so any input at all is greatly 
appreciated!

According to our manager, the physician signature is for clinical testing only, 
not anatomic pathology.  If anyone knows that this is inaccurate, would love to 
see the written rules!!
