[Histonet] Leica 2135 parts

I am looking for a flex shaft cable for our Leica microtome 2135. I have been 
told by Leica they no longer service or carry parts for this machine any 
longer. Ours is in excellent condition and a new one is not in our budget. Is 
there a Leica “junkyard”?

arrival condition and took pictures. I took a small piece and processed it last 
night and will be cutting it in a few minutes. I am pretty sure the results

Are you referring to a whole heart? If that is the case I think the fixation of 
the tissue will be very poor because Karnovsky's penetrate very slowly.
I do not think it will be worth the effort and materials to process this tissue.

paraffin processing.

solution from another lab and voila, perfect staining. Thank you to everyone

Hi Peggy,
No, this is a batch I have been using since 12/12. I purchased it from Sigma. 
It seems faint after the ammonium hydroxide . I did a run yesterday and I did 
get some blue, but faintly, not as bright as I am used to seeing. Everything 
else was picture perfect. Ray suggested that it may be a change in my tap 
water. I have our current report and am waiting on the report before that one. 
I am doing another run today and am considering rinsing in DH2O after the 
Alcian Blue and ammonium hydroxide.
What do you think?

[Histonet] harder paraffin

Hi Histonetters,
Good Friday to you all. Do you know if there is a “harder” paraffin  that may 
offer more support to the tissue while cutting? I currently use Paraplast Plus 
and love it, but now I am cutting some polymer material and there is some 
tearing and was thinking that maybe a different paraffin would help. I am 
trying to avoid embedding in plastic.

[Histonet] Zinc-Tris mouse hearts

Hi,
I am going to have some mouse hearts that have been fixed in zinc-Tris  
fixative. I have never worked with this before. My question is about 
processing. After doing some looking, I found an article that advised checking 
the solutions on the processor , the first couple of alcohols , to be sure 
there is no carry over of formalin. I understand that the object if this 
fixative  is to avoid formalin but is this step necessary?

Hi all,
I have a couple of questions regarding decal and staining. I have done some 
searching but could use some hands on experiences.
It involves bone regrowth. The sample will vary in size. From my reading, it 
seems that 5% formic  or 10% EDTA is preferred. The researcher asked about 
acetic acid, but I have never heard as acetic alone as a decal, isn’t it just 
used to balance pH of the EDTA? It is my understanding that the decal step will 
be done somewhere else then the sample sent to me. That lab feels that HCL 
decal will not  not affect the sample. I think otherwise. The staining they 
have talked about is the Goldners Trichrome mainly. The researcher mentioned 
Safranin O but from my conversations I think they have little knowledge of 
histology and I work in a cardiovascular lab with my only experience has been 
with mouse tibia and these samples will be from a larger source.
Type 2 collagen was brought up but when I said that was an immuno stain they 
did not realize that.
Si in a nutshell, what decal sol. would be better and what stain?

I don't know of any modifications for Eager's but there are modifications for 
the Fink-Heimer technique as well as the Nauta-Gygax technique that can be used 
on paraffin-embedded tissue. I've only come across these in passing and you may 
have to do a little digging to find a protocol.

 preferred. However, in my experience, I've come to find the Bielschowsky 
 method very useful for staining nerve fibers, but I would not consider it so 
 for demonstrating axonal degeneration. I would suggest that you maybe 
 consider Eager's method for degenerating axons. Unlike Bielschowsky's, 
 Eager's actually distinguishes those fibers that are normal from those that 
 are degenerating.

I had this problem awhile back. Did you change your solvent for 
deparaffinization? I changed from xylene to another solvent and realized I had 
to extend the time my slides were in it.

[Histonet] frozen lung sections

Hi all,

 I have received some lung samples on OCT. These lungs have not been in
PBS or any fixative. They went directly from dissection into OCT and
frozen. 

 I cannot get a section. The temp of the cryostat is -20. I have done
some reading and it seems the best results come from immersion in PBS,
then 4% paraformaldehyde then into sucrose. Can anyone clarify the
procedure or offer advice? I am unfamiliar with this technique since
most of my frozen sectioning is heart tissue and it does not present the
same problem as the lung.

[Histonet] staining questions

Good morning!

 I will be staining vessels for proteoglycans using Alcian Blue. I was
going to use the 2.5pH  protocol with a Nuclear Fast Red counterstain.
Does anyone have any other suggestion? The investigator does not want
Movats.

 Also, I normally use PTAH as my fibrin stain for clots but was thinking
that there may be something better. I found the Fraser-Lendrum but the
protocol stated that Zenkers was the best fixative and that is out of
the question. So again, any suggestions? Thank you in advance.

[Histonet] Movats on mouse aortas

Hi,

 I have had a problem with splotchy elastin staining on certain
sections of mouse aorta. I use the Modified Movats and do not use a
microwave. But from this one lab  some areas of the elastic fibers will
retain the hematolylin after differentiation while some areas lose it
completely, all in the same section. 

 They fix in 10%NBF and the processing gets done at their lab and I
receive the unstained slides. My control does not have this problem.

Any ideas?

This had nothing to do with an inspection, but follows this comment. There were 
3 plants in our lab and there was one histotech who starting sneezing and got 
congested whenever he sat at his microtome. The plants were in front of him on 
a raised shelf. Finally we had a thought it may be the plants so we took them 
down and had a closer look and indeed there was a moldy material growing in the 
soil. Removed the plants and it was resolved.

Hi All,
I think all of you are missing the point of Patti's question.  She stated that 
her lab was dinged for having plants in the lab by a CAP inspector.  
I had the same thing happen to me years ago.  The inspector stated that plants 
attract insects that can contaminate a supposedly clean environment.  
Patti has an extremely well organized lab that only had a small phase (1) 
deficiency last year.  I think the inspector couldn't find anything, so they 
had to come up with this ridiculous infraction.

I don't know Sally, but where I worked many moons ago I had a spider plant that 
was extremely prolific.  The powers that be made me remove it from the lab for 
an inspection.  It went to live in one of the administrator's office for 
several months.  And died!  I think it needed the fumes!  Or it missed me.

I think it's the fluorescent lights that makes them thrive.  The
absorption of Fume Matter is a secondary, but beneficial, effect.  You

[Histonet] contrast

Hi,

 I have some mouse hindlimbs that have catheterized with Omnipaque. The
Omnipaque contains 302 ml of iohexol which is equivalent to 140 mg of
organic iodine per ml. The mice will be sacrificed immediately after
being radiographed. They would like them to be processed in paraffin. I
am wondering if this will have any effect on the muscle.

I don't like the generality of this statement. I work in a research lab and my 
stuff is not crap. The people that I work with do not produce crap, HT or no 
HT. I find research more challenging in fact than clinical. Everyday I get 
animal specimens of all types in various fixatives. Some have device implants 
of various materials and some are looking for cellular changes. Everything must 
be of photographic quality. I take great pride in my work and other techs in my 
department. 
 At times I do receive samples that have been abused by the doctor or 
research fellow but that is not my labs' fault and we will speak to the 
researcher about future handling of their samples.
Maybe I misunderstood and am reacting too strongly to you statement, but I get 
tired of some thinking that research histo techs are second class citizens.

Ever since I started work in a lab in 1957, salaries have been poor and not
kept up with cost of living or reflected the skill of many of the
histotechs.

I feel that one of the major problems is that we have several categories of
histotechs.
Certified (I mean that in the nicest sense of the word!).
Certified and also highly skilled.
Skilled via on the job training.
Robotic technicians.

Until a system is put into place that requires all histotechs to be
certified, or in training along a specific career pathway then things will
never change. Along with this must be a system that requires employers to
have well defined career paths for all their histotechs. This must include
training outside of the routine work that they need to do in their specific
laboratory.
I must agree that getting rid of the practical part of the examination was
how can I put this delicately ... imbecilic.
The workplace needs to get back to being less of a bottom line factory in
which careers of the employees is often not a consideration to a place where
career paths are an integral part of the employment. The optimal environment
is one where there is a two way street where both employer and employee
benefit.
While money is a consideration, it is not the only reason that individuals
continue to work in a specific lab.

the other Old TEchs that came through the hard way like I did- it was much
more of a challenge and the certification really meant something you worked
HARD for!
How do we pass that on the our newer techs. I think it is up to us to make
sure they know how to do things- how to troubleshoot- how to make things by
hand should they not get in the order of pre-made stuff!

I was referring to those researchers that bring the crap (non-techs trying
to do the histo). Not what we do. I do feel we are second class citizens
because it is felt that anyone can do histo. They read a procedure in a
paper and think we can do magic or, my favorite, give us IHC conditions that
are done by hand and we do it on the autostainer.
Sorry if you took it the wrong way. I was not picking on histotechs, but the
non-knowledgable researcher that think they are histotechs and know more
about it because the might have a PhD. I could get really started on that. I
had to explain to someone the difference between a cross, longitudinal and
tangential section and this person was supposed to have a science
background!

descriptive protocol and all the protocols in internet are already
assuming that you have some plastic embedding knowledge and I have never

and simple to program.

Try to get a demo from Sakura.
Regarding glass vs. film:
 
FILM: faster and slides can be filed immediately; after YEARS the film may 
become loose, but only if it was placed with less than needed xylene.
 
GLASS: better for photomicrography IF you use apochromat high power dry 
objective and glass remains better in the long time. They have a higher failure 
rate (more interruptions in the work flow). Sometimes slides need to wait flat 
for days before being filed.
 
I personally prefer film, the net result outweigts its drawbacks.

[Histonet] wavy mid myocardium

Hi,

I processed some mouse hearts that were in Z-Fix for 24 hrs. then put in
70% for anywhere from 24-48hrs. They were processed on a short run. The
epicardium  looks good but the mid myocardium is wavy.

15 min 80%

15 min 95 x 2

15 min 100% x 2

15 min Xylenes x2 

10 min paraffin x 4

Any suggestions?
