[Histonet] Leica 2135 parts
Hi, I am looking for a flex shaft cable for our Leica microtome 2135. I have been told by Leica they no longer service or carry parts for this machine any longer. [Histonet] Karnovsky's
Hi, I received a heart that was fixed in 2% Karnovsky’s. I was told it was initially kept under refrigeration but then stored at RT. I do not know how long the heart has been stored at RT. By the look of the tissue I would say quite a while. Is this tissue compromised in any way? Should I remove it from the Karnovsky’s and store in in something else? Good Friday to all. Histology 101. The Alcian Blue dye itself had lost its efficacy. I borrowed a solution from another lab and voila, perfect staining. Thank you to everyone for their suggestions. Hi Peggy, No, this is a batch I have been using since 12/12. I purchased it from Sigma. It seems faint after the ammonium hydroxide . I did a run yesterday and I did get some blue, but faintly, not as bright as I am used to seeing. Everything else was picture perfect. Ray suggested that it may be a change in my tap water. I have our current report and am waiting on the report before that one. I am doing another run today and am considering rinsing in DH2O after the Alcian Blue and ammonium hydroxide. What do you think? [Histonet] harder paraffin
Hi Histonetters, Good Friday to you all. Do you know if there is a “harder” paraffin that may offer more support to the tissue while cutting? I currently use Paraplast Plus and love it, but now I am cutting some polymer material and there is some tearing and was thinking that maybe a different paraffin would help. I am trying to avoid embedding in plastic. [Histonet] Zinc-Tris mouse hearts
Hi, I am going to have some mouse hearts that have been fixed in zinc-Tris fixative. I have never worked with this before. My question is about processing. After doing some looking, I found an article that advised checking the solutions on the processor , the first couple of alcohols , to be sure there is no carry over of formalin. I understand that the object if this fixative is to avoid formalin but is this step necessary? Thank you in advance. I have done some searching but could use some hands on experiences. It involves bone regrowth. The sample will vary in size. From my reading, it seems that 5% formic or 10% EDTA is preferred. The researcher asked about acetic acid, but I have never heard as acetic alone as a decal, isn’t it just used to balance pH of the EDTA? It is my understanding that the decal step will be done somewhere else then the sample sent to me. That lab feels that HCL decal will not not affect the sample. I think otherwise. The staining they have talked about is the Goldners Trichrome mainly. The researcher mentioned Safranin O but from my conversations I think they have little knowledge of histology and I work in a cardiovascular lab with my only experience has been with mouse tibia and these samples will be from a larger source. Type 2 collagen was brought up but when I said that was an immuno stain they did not realize that. Si in a nutshell, what decal sol. would be better and what stain? Sorry for the rambling. However, in my experience, I've come to find the Bielschowsky method very useful for staining nerve fibers, but I would not consider it so for demonstrating axonal degeneration. I would suggest that you maybe consider Eager's method for degenerating axons. Unlike Bielschowsky's, Eager's actually distinguishes those fibers that are normal from those that are degenerating. [Histonet] frozen lung sections
Hi all, I have received some lung samples on OCT. These lungs have not been in PBS or any fixative. They went directly from dissection into OCT and frozen. I cannot get a section. The temp of the cryostat is -20. I have done some reading and it seems the best results come from immersion in PBS, then 4% paraformaldehyde then into sucrose. Can anyone clarify the procedure or offer advice? I am unfamiliar with this technique since most of my frozen sectioning is heart tissue and it does not present the same problem as the lung. [Histonet] staining questions
Good morning! I will be staining vessels for proteoglycans using Alcian Blue. I was going to use the 2.5pH protocol with a Nuclear Fast Red counterstain. Does anyone have any other suggestion? The investigator does not want Movats. Also, I normally use PTAH as my fibrin stain for clots but was thinking that there may be something better. I found the Fraser-Lendrum but the protocol stated that Zenkers was the best fixative and that is out of the question. So again, any suggestions? Thank you in advance. [Histonet] Movats on mouse aortas
Hi, I have had a problem with splotchy elastin staining on certain sections of mouse aorta. I use the Modified Movats and do not use a microwave. But from this one lab some areas of the elastic fibers will retain the hematolylin after differentiation while some areas lose it completely, all in the same section. They fix in 10%NBF and the processing gets done at their lab and I receive the unstained slides. My control does not have this problem. Any ideas? There were 3 plants in our lab and there was one histotech who starting sneezing and got congested whenever he sat at his microtome. The plants were in front of him on a raised shelf. Finally we had a thought it may be the plants so we took them down and had a closer look and indeed there was a moldy material growing in the soil. Removed the plants and it was resolved. Hi, I have some mouse hindlimbs that have catheterized with Omnipaque. The Omnipaque contains 302 ml of iohexol which is equivalent to 140 mg of organic iodine per ml. The mice will be sacrificed immediately after being radiographed. They would like them to be processed in paraffin. I am wondering if this will have any effect on the muscle. Thanks. [Histonet] wavy mid myocardium
Hi, I processed some mouse hearts that were in Z-Fix for 24 hrs. then put in 70% for anywhere from 24-48hrs. They were processed on a short run. The epicardium looks good but the mid myocardium is wavy. 15 min 80% 15 min 95 x 2 15 min 100% x 2 15 min Xylenes x2 10 min paraffin x 4 Any suggestions? Thanks.
