Hello all,
The School of Veterinary Medicine at the University of Wisconsin, Madison has a 
full-time position for a Necropsy Technician. The position is responsible for 
assisting the veterinary pathologists in the biopsy and autopsy service - much 
like a human hospital but with a much wider variety of species from the 
veterinary teaching hospital, (dogs, cats, horses, cows), the DNR, (wolves, 
deer, raccoons, birds), and the Milwaukee County Zoo, (fish, primates, kudu, 
rhinos, penguins, and many other exotic species. I'm not gonna lie; It's pretty 
cool.
This is a terrific group of co-workers and pathologists who are dedicated to

[Histonet] Block Holder for Leica Rm2255

The handle snapped off our RM2255 microtome block holder, and 
the replacement we were sent does not open the clamp far enough. (We are 
currently managing by using an allen wrench in the drilled out hole...) Does 
anyone have any suggestions?

I'm looking for a deep water bath, (3" minimum), that has an 
interior light source. Most of them seem to be only 2" tall, or have a slanted 
front control panel that none of us in the lab want. Does anyone know of a 
brand of flotation water baths that are 3" deep and have interior lighting?

[Histonet] Jeepers Creepers, Coverslipping Peepers!

Hello all,
Does anyone have a source for oversized coverglass? (35x50, 
35x60, 45x50, 45x60) We section a lot of globes from the ophthalmology lab, 
some of which are from horses, cows, etc. that are put on the larger slides. 
Fisher used to carry their own brand, but it's apparently discontinued them, 
and their "alternate" product is $50/oz!

[Histonet] Thyroglobulin TGB04+TGB05 Not Working in Canine and Feline

We had to switch sources (to Novus) for Thyroglobulin 
TGB04+TGB05 and cannot get it to work in canines or felines. Data sheet 
suggests 1:100, pH6 citrate antigen retrieval. We've also tried zero AR and pH9 
EDTA AR, to no avail. Any suggestions?

I have 1 inch thick cross sections (band saw) of calf hooves. 
The pathologist has already decaled the bone, and now we have to soften the 
hooves so she can take thinner sections for processing.


* Which is better, potassium hydroxide or sodium hydroxide, and what 
concentration?

* How often should they be checked for softening?

* What is the relative air speed of an un-laden swallow? (European)

We currently cut the MUM-1 fresh, and the Synaptophysin. (We seldom do a 
Syanpt., and have found the fresh cut patient tissue has been positive while 
the not-so-fresh positive control slide is negative.

[Histonet] Ortho-keratotic Mineralizing Odontogenic Cyst

Hello all,
Does anyone have any ideas on de-mineralizing an 
ortho-keratotic, mineralizing, odontogenic cyst? It is so hard the pathologist 
cannot cut it into small enough pieces to fit into a cassettes. (It's been in 
decal for days with no change.) I was thinking of taking one of the chunks and 
putting it in potassium hydroxide under the hood, and monitoring every half 
hour or so. Anyone have any other ideas?

Our Sakura tape coverslipper is doing something I've never 
encountered before - the lever that holds the blade and goes up and down and 
cuts the tape, has gotten sticky and slow. It will chop down fine, but has a 
hard time returning to the up position. Before I take the back off, I'm just 
wondering if this has happened to anyone else, and if so, how it was rectified.

[Histonet] PT Module Spare Part

reagent level sensors? (The black bar on the back of each tank.) Our pH 9 AR

We are in the market for a new IHC stainer. (Our Lab Vision 720 
is no longer reliable.) We average about 20 slides a day, but sometimes get 
research projects that are 50+ slides.  Not interested in a closed system; it 
is for a veterinary histology lab, and we need the flexibility to tweak 
antibodies for dogs, cats, horses, and zoo animals. I'd really appreciate 
feedback from users who are in a similar lab setting.

We are having difficulty sectioning mouse tissue, (brain, 
spinal cord, liver, and spleen), on paraformaldehyde fixed tissue. Has anyone 
had issues with paraformaldehyde fixed tissue? They were processed routinely, 
starting in 10% NBF, with other tissues, and we are cutting them at 3u.

[Histonet] "Supa Mega Clamp"

We use the "Supa Mega Clamp" (Ted Pella) for sectioning large 
tissues in the oversized cassettes. Lately, it seems not to grip as tight as it 
did when we received it a month or so ago. The literature is little more than 
photos of how to attach the clamp to our microtomes. Has anyone experienced 
this? Is there an adjustment that can be made?

[Histonet] Pathology Band Saw

Hello,
Can anyone recommend a band saw for pathology? We are looking 
for a new band saw for use in necropsy and research. It has to be a "meat" band 
saw, as workshop band saws aren't compatible with wet tissue.
A table-top model is preferred, and cheaper than a floor model. (Plus, the 
floor models all seem to come with a sausage-maker, which is pretty gross.)
If you have used a table-top band saw in your pathology lab, 
I'd appreciate hearing from you.

[Histonet] Refurbished Leica RM2135

Hi everyone,
I have a newly refurbished Leica RM2135 at my disposal, but the 
knife holder is for high profile blades. 2 questions.
1-Is it that different using high-profile blades vs. low. (I have used 
low-profile blades forever.)
2-There is a slot on the bottom of the blade holder, and I seem to remember 
that it can be converted to a low-profile holder by sliding an narrow metal 
strip into that slot. I know I've done this before, but it was long ago, and 
the used equipment guy says there's no such thing.

[Histonet] Microm HM 355S Issues

Hello,
We have 2 Microm HM355S motorized microtomes. We use the motor 
to move the block close to the blade, but turn the wheel manually when 
sectioning. Both are "jumping" forward on their own occasionally while 
sectioning, which is unsettling and chunks into the block. One of them also 
backs up on its own as you're manually turning the handle, just as you're about 
to get a ribbon.
The stepper motor has been replaced on one with no improvement. 
Any suggestions, remedies, or tales of similar issues and how they were 
resolved would be most appreciated.

Picasa, the program we currently use for re-naming and 
organizing our gross digital photos, will no longer be supported by Google. If 
anyone can suggest another program for this purpose, please let me know. We 
don't need any fancy editing of the photos themselves other than cropping and

Wisconsin, as well as many research needs at the School of Veterinary Medicine. 
We are a full service lab providing processing, embedding, microtomy, 
histochemical and immunohistochemical staining.

[Histonet] Hematoxylin Precipitate

Hello all,

Has anyone using Richard Allen Hematoxylin-2 noticed an odd 
artifact on the slides after using the Hematoxylin for more than a few days on 
their stainer? We are seeing small spore or pollen-like blue dots here and 
there on the slides. It is not coming from the water bath or our water supply 
on the stainer. I used sterile gloves, opened a new case of slides, dipped them 
in DI water, then in the RA Hematoxylin 2 on the stainer, then in DI again, 
air-dried and coverslipped them, and the blue dots were there. The only way we 
got rid of the blue artifact was to use new RA Hematoxylin-2 every 2-3 days, 
which is a bit expensive.

Thanks for your input, and if you can recommend a different, 
reasonably priced hematoxylin, that would be awesome.

August 15th. (One of my techs had a bicycle accident and broke his collarbone 
among other things, and there are several vacations coming up for other staff 
as well.)

haven't needed to use one since I worked in Anchorage, and that was last 
century!

[Histonet] IHC Buffer (wash)

Hello everyone,

Got your Christmas shopping done? (Didn't think so...)

Anyway, I'm looking for an IHC PBS/Tween buffer wash concentrate that is 
comparable to the very expensive Lab Vision PBS/Tween buffer wash. I see some 
at VWR and FISHER (no shipping charges for our lab) that are 0.5% and 1% Tween 
20 washing buffer in PBS.

Which percentage is best for IHC on a Lab Vision stainer using 
a polymer detection kit? As usual, your input is appreciated.

[Histonet] Issue with Henry's Wonder Wax Enhanced

Hello fellow Techs,

Has anyone noticed a change in the Henry's Wonder Wax, 
Enhanced? (Used for embedding, not infiltrating.) We have noticed that it is a 
lot stickier when we are sectioning. (To our picks, forceps, and fingers.) We 
have checked our temperatures, and all is well. I have asked the company, but 
they have not replied.

Can anyone recommend another embedding paraffin? (We switched 
from Paraplast Plus about a year ago when we noticed changes after the original 
manufacturer was bought out.)

[Histonet] Parts for Sakura Tissue Tek 5 Embedding Center

Does anyone know of a used Histology equipment company that 
sells Sakura parts? We have a fairly new embedding center, (but past warranty), 
that needs a single heater unit. Sakura only sells the entire dispenser unit 
for nearly $8,000. I don't mean to offend any Sakura fans, but I don't think 
that's right.

[Histonet] Leica-Surgipath EM Paraffin Issue

Has anyone else noticed a change in the EM400 embedding medium from Surgipath? 
We have not changed anything in our embedding, but now the EM400 results in 
cracks in the blocks. Leica says although the labeling has changed to indicate 
the EM400 is now also for infiltrating, the paraffin has not changed.

[Histonet] CD20 suddenly has high background

We do CD20 on canine and feline samples and are suddenly getting lots of 
non-specific background. We use:



CD3, F7.2.38 from DAKO diluted 1:400

LSAB+ and DAB+

Avidin and Biotin block



I even tried it with a polymer detection kit, but to no avail. Has anyone else 
had recent difficulty with this T cell marker?
