We may opt to have the housekeeping crew put on a new coating that supposedly

We have had reasonable results based on rapid thawing of frozen samples in room 
temperature buffer followed by formalin fixation or rapid freezing in OCT.  
With such large samples, though, there are bound to be large ice crystals in 
the interior of the specimen regardless of the method used for freezing.

Hello All,

I have compiled all of the responses to date regarding the question I posted on 
histonet about too much volume and reverse capillary action: (I've changed font 
styles to differentiate between responses)

I have used them for quite some time.  I was having problems with uneven 
staining and determined that it was from the being too full and was drawing up 
liquid from the bottom.  There is a small opening on the end and I just empty 
the solution from it.  I usually empty it after rinsing the antibody off and 
then usually before putting DAB or DAPI on depending on the number of slides 
stained.  As for washing, I just wash in a solution of soapy water and then 
rinse in distilled.  I use the coverplates for several staining times.  I love 
the sequenza and prefer it to having an automatic stainer.


I still use these and have 9 racks. 
The coverplates can be found on eBay, but I clean them in soapy bleach water, 
rinse well, and dry until the plastic gets brittle and they break.
I use AEC 99% of the time so I do not generate the waste of DAB.
If the racks are filing up with reagents and solutions, leave an empty slot and 
pipet it out. Otherwise, I dump it and and rinse after the run.
For the antibodies and detection solutions, you only need 3-4 drops. I only 
fill the well to the top with rinses.


We use these units, I have never had an issue with the waste reagents touching 
the bottom of the plates.  We never use more than 3 drops of kit items, the 
wells are filled with buffer.  It never seems to be alot of waste.
After we finish staining we just rinse the holders out with running water for a 
few minutes and let air dry.


We  empty them before they get to that stage, as this  would be  bound to 
affect the  efficient working of the Coverplates.


This is Tyler Liebig with the Thermo Scientific IHC group.  I'm responding to 
Keri's questions regarding the Sequenza manual staining.  Keri you are correct 
that if the waste is not emptied and builds up too much in the bottom of the 
sequenza unit it will definitely stop the capillary action (Reagent can't flow 
both ways :-).  However, this can be avoided by emptying the waste regularly 
between runs.  

You bring up a good question though that I didn't know the answer to: How many 
slides can be stained before the waste hits the slides? I did a quick 
calculation below that I hope is helpful.  

Total Waste Volume of Sequenza Rack:
In a Sequenza rack the slides are held about 1.5 inches above the bottom of the 
tray.  This allows about 270ml of waste before the level is close touching the 
slides.  

Reagent use per slide:  If you stain a single slide with a long protocol then 
worst case scenario is about 7 steps.  (H2O2 through Counterstain with a two 
step polymer)
Volume of rinse buffer: This means you would rinse 8 times 2ml each (16ml total 
rinse buffer per slide worst case).
For the staining reagents: The recommended volume is 0.1ml per reagent and 
0.5ml for chromogen.  So for 6 reagents plus chromogen that is 1.1ml and I will 
double it because I know some people add way more than needed.  So 2.2ml per 
slide worst case.

So to sum it up worst case scenario 18.2ml of waste is created per slide which 
means 14 slides (270/ml/18.2ml=14.8) can be stained before the waste risks 
touching the slides.  The rack holds ten slides total. 

In the end I recommend users play is save and empty the waste after each batch 
of slides stained even though most user generate much less waste than the 
calculations above.

I just wanted to say thanks again to all of those on histonet.  The questions 
that are asked go a long way in teaching troubleshooting.  Also, a few weeks 
ago the question was asked about purchasing forceps. I don't remember when or 
who.  One of the suggestions was jewelry forceps from craft stores such as 
Hobby Lobby.  That was a great suggestion!!  I took a field trip to Hobby Lobby 
and found some nice curved forceps with ergonomic finger pads for $3.99.  
Instead of ordering from Hobby Lobby, I called the distributor and was able to 
get 15 pairs for $2.99.  The tension and the weight of the forceps are good, 
and have a non-stick coating.  We will be placing them in our microtomy kits 
for next year.  For those interested, I also found Collins Scalpels (paraffin 
knives) at D R Instruments for $6.45 each if anyone needs some.

This was a good discussion and a some little rough comment  but we learn from 
it. I have read all the feedback on xylene and its substitute and it  gives us 
a lesson to become more better histotechs. Just this morning one of our 
administrator passed by near our processor while my co worker has just opened 
the processor retort for cleaning and smelled xylene in the air. He stopped and 
asked us why he was not using a mask while doing that. I replied that was only 
a minimal amount of xylene in the retort that was left after the cleaning cycle 
and he said well I just want to make sure of your safety. When we are so used 
of the smell sometimes we do not think of our Safety and this discussion taught 
us again to think first of our own Safety whether you are sensitive or not to 
xylene or any chemical.
To shelly, I wish your employee the best and all success in the field of

One point to bring up, and I am sure that Leica service will do this, is the 
brand of knives you are using. Different brands have slightly different 
thickness. This is not related to the height of the blade. We have found that 
we need to use the blades that are used by our service company to calibrate the 
microtomes to have the correct tension in the holder and not any problems with 
the quality of the sections.

Thanks for all of the Microtome feedback, it has been very helpful.  It sounds 
like everyone that has the RM 2235 loves it, and are not having a lot of 
problems.  Our Leica RM 2235 has problems with blade holder tension. We worked 
with Leica when it was just about 14 months old but they were no help. The 
problem is the front portion of the blade holder that clamps the blade in place 
is too thin or just not made of strong enough material. We don't have much 
tension on the blade while sectioning but the plate starts to curl or bend at 
the top where it meets the blade. Then the blade tension is inconsistent and 
the sectioning is inconsistent. We have been using a Leica 2030 blade holder 
for the last few years but its a little too short. Any suggestions on a blade 
holder  Has anyone had similar issues with their RM 2235

It must be out of warranty now, but why don't you price a new blade holder? Or 
better yet, ask Leica if they would replace the original one for you? If you do

Thanks to all for your suggestions for cutting cross sections of
cortical bone. I have passed them along.

Now, what is the latest on CD3 IHC in FFPE rat tissues. I saw Ray
Koelling's suggestion in the archives using an Ab from BD Biosciences.
Is it cat # 550295?

What others if any do you suggest? HEIR ? protocol tips?

Dako has a rabbit polyclonal that works in rat.  The other thing with
the bone, is place the slide with a section on a flat hotplate (60 C)
until it turns clear - not too long or the articular cartilage will
flip.

I was able to speak to Larry M.  about  my tissue processor problems.  thank 
you all for your  help.  

I am now looking for perhaps a broken MVP-1 or 2 since I am no longer  able to 
get the CRT display parts to fix mine.   they are  interchangeable and if I 
could find a discarded MVP I could get mine running  again.   Anyone have a 
mothballed one sitting in the basement?

are there any useful histological techniques to stain capillary/blood vessel in 
brains?
and permit co-immunostaining of other cell markers?

PAS will stain all connective tissues, especially the myelin, thus you have to 
interpret your data well?

[Histonet] thanks Henwood for the PFA discussion

At my previous job,I got so tired of the PFA discussion with a few folks
that I would hand them two containers of NBF one labeled PFA and one
labeled NBF and tell them to test it and they always went for the PFA.
I mean same stuff!  All in their heads man.  So those folks always got
the special labeled container.  Little placebo action never hurt eh?  I
notice that recycled formalin seems to boil off methanol too which is
what I use mainly- recycled.  Sometimes I have to add a little full
strength, but seems like it reads less alcohol after I run it thru the
recycler.  I see the billing point of methanol is 64.7 so it would
appear to be knocked off.

[Histonet] Thanks for help on desperately seeking help for cryosectioning and further question

Dear All,

thank you so much for your suggestions on improving my cryosectioning 
results. I tried several of your suggestions, however, the quality of my 
sections did'nt improve so much.
I get perfect sections as long as I only cut the OCT embedding media. As 
soon as I reach embryonal tissue, only the tissue starts to tear up. In 
some of the embryos I loose almost all tissue. In my opinion this can 
only be caused by the fixation of the mouse embryos.

Here is our protocol:
1. Fixation in 4% PFA overnight at 4°C
2. 30% sucrose solution at 4°C till embryo sinks to bottom of the tube
3. transfer to 1:1 mix of 30% sucrose solution and OCT for 4h at 4°C
4. transfer to 100% OCT and freeze quickly in isopentane (cooled down 
with dry ice for approx. 30min before use).

5. store at -80°C

Is there something wrong? Should I follow a different protocol`?
How long can you store OCT-embedded tissue? Are 6 months too long?
