Hi Jamie,
1. The sheets have adhesive part and holding non-adhesive part. The sheet
is as big as a regular A4 size. The length of the individual tapes is fixed
but you can cut the sheets in different widths depending on the width of
the sample. You can also order sheets with different fixed lengt
Hello Dear Friends,
Our lab really likes the light sky blue color of Ventana Hematoxylin (as
opposed to other more purple ones). I noticed the million types of
Hemtatoxylins in the "Theory and Practice of Histotechnology" book. Does
anybody knows a Hematoxylin that will give this blue color?

Hello Histonetters,
I have read in the past e-mails that some of you have used CD8a Rat
monoclonal from Angio-Proteomie cat#mAP-0036 for IHC on FFPE mouse tissue.
I have tried the antibody on Lieca Bond RX and did not get any good result.
Could you share your protocol?

Dear Histonetters,
I need your help to find good antibodies for IHC on mouse and human FFPE
tissues like normal tissues and tumors. Please share any information you
think might be helpful.

Dear Histology Experts,
I need your help. I am trying to stain undecalcified bone (adult mouse
limb) with Alizarin Red S.
I was thinking about two options:
1. Fixed frozen sections with Cryojane tape transfer system then staining
the sections with Alizarin Red.
2. Alcohol fixed whole limb (whole
Hi Anna,
Your subject line is F4/80 and you are asking for CD68. I thought they are
different antibodies, but I may be wrong.
We have never been able to find CD68 ab that works well of FFPE mouse
tissues. Recently we had success with F4/80 Rat anti-mouse Clone: BM8 from
BioLegend. It only worked w
Dear Histonetters,
I wonder if anyone has done myeloperoxidase stain on FFPE or frozen lung
sections?
I would appreciate any help.

Hi Elizabeth,
I always read and benefit from the messages here. Now I so happy that I
think I might be of some help to you or also others.
When I do enzymehistochemistry to detect the beta galactosidase enzimatic
activity using x-gal as a substrate, I mostly use fresh frozen tissues and

Dear Histonetters,
I wish you happy holidays and also would like to know if anybody has good
antibody for FFPE rat, mouse or human tissues for FOXO1 or FOXO3 ( also
pFOXO1 or pFOXO3 would work)?
Thanks,

I have 2nd and 3rd edition of Freida Carson's Histotechnology-A self
Instructional text book. When I read the : 2nd edition pg 98, or 3rd
edition pps 117-118. I noticed that this is not for OilRed O staining but
for forozen sections in general.
When I read: Freida Carson's Histotechnology-A self I
HistoDeck questions are companion of the Carson' s book. Carson fixes in
37-40% formaldehyde for OilRed O.
Any acetone or alcohol will dissolve the fat and OilRedO will be negative
or suboptimum.
The answer should be "A"

Hi All,
Does anybody have a protocol for Gram stain on FFPE mouse intestine
sections. We are looking to distinguish between Klebsiella and Vancomycin
Resistant Enterococcus (VRE). I would like to avoid using Picric acid as
some protocols suggest.
Thanks in advance.

Hi All,
Actually we have tried the new lots of the dead goat antibody with Ultamap
kit (polymer) on Ventana and worked pretty well. But the kit was so
expensive and we did not continue.

Dear Histonetters,
I do not think there is a good CD31 (PECAM-1) antibody that will work on
FFPE mouse tissues. If you have one please let me know. We use CD34 rat
mono (Abcam) and Meca-32 Rat mono (Dev Hybridoma Bank Iowa Univ.) to label
endothelial cells of blood vessels in FFPE mouse tissues.

Hi Collen,
You had great responses to your question. I do stain mouse femurs for IHC
and IF. We fix in NBF overnight and decal for 4-5 days in 0.5M EDTA pH=8 at
room temp. After decal we wash with running tap water and re-fix for 4 hrs
with NBF. 90% of the antibodies we use on undecal tissues work
Dear Histonetters,

Dear Histonetters,
I am going to do Immunofluoresence staining (beta-catenin, Notch1 and

In the literature I have encountered different protocols for removing the
MMA.

Hi Maria,
Often people freeze the tissues right after dissecting them out of the
animal because either they are not sure what to do or they do not know
better.
Then they bring the frozen tissues kept at -80 C freezer and request
paraffin embedding.
I usually prefer to thaw them gradually. I place
