Make it simple if you like just remove the vacuum tube from the carb/boot so you’re left with 4 nipples. What’s your bike, a CB650SC?

Have you had it running bypassing the gas tank? I wouldn’t have suspected the float valve or needle valve. I suspect the pilot jet or slow speed jet (same thing) but if you’ve cleaned it then I would suspect some sort of contamination prior to the fuel getting to the carbs. Is the spark plug in #4

#4 cylinder would eventually get hot with the bike running on three anyway. Your hand probably can’t tell the difference between 350 degrees and 180 degrees.
