Stefano, to run SA, you will need a server running some form of MTA (Mail
Transport Agent) such as Sendmail, Postfix, Qmail, etc

bridge does:
check messages
clean viruses
change header
change body
notify users
Layer 2 protocol are things like: ethernet, PPP, HDLC, STP... SpamAssasin, or any is not aware of any layer 2 protocols. Niether is any antivirus program I know about.

On linux or BSD, you can easily set some bridge, at layer 2 level, and have it analyse all packets, with some netfilter/ipfilter/whatever rule to handle locally SMTP/whatever traffic.
This even sounds like a very good idea,
