Anyone knows a gameboy emulator called no$gmb? I've contacted
its author and presented the very same question which was turned
down by nintendo to him. I think that you might find the answer
quite informative:

No, it isn't rs232 compatible. The port has a clock signal, and two data
signals (for sending and receiving). Every time the clock signal gets
high (or low, not sure), one data bit is send (and read).
The clock signal is controlled by the 'master' gameboy, as long as you
let the PC control the clock signal (and make the gameboy the slave
computer), it's no big deal to connect it via LPT port.
The gameboy internal transfer speed is 8kbits/second, but you could use
faster (or slower) clock speeds if you generate the clock signal on your
own.
