Depending on how confident you're feeling, one of the best things you
might want to tweak is using the binary drivers for your graphics card -
try following the instructions at:

https://help.ubuntu.com/community/BinaryDriverHowto

It certainly made the desktop on my machines (nVIDIA GeForce 5700 and
ATI Radeon 9600) much more responsive.

You might also want to try installing the kernel packages for your
particular processor (in Synaptic there are various ones for i686, AMD
K7, etc. which are more optimised than the standard i386 ones). It's
usually a simple matter of using Synaptic to install them, and then
rebooting.
