VBoxManage clonehd <existing vm name> <new vm name>

Check the User Manual - there are some other features to clonehd.

I just did this to clone a WinXP vm from a 3.2.8 VBox to test it with 4.0 VBox on another system. After you clone the vm, you need to create a new VM (e.g., using the GUI), but use "Use existing hard disk" instead of creating a new one. The existing one is the clone you just made.
