I have experimented some more with limited success. Oddly enough, the combination works on rare occasions, but I am not sure what determines this. I used option command shift x, as well as a few others, to make the start speaking text shortcut. Other ones I have tried have not worked, so I am wondering if it is a system specific problem. Could it be that Spark is installed? As far as I can tell, btw, none of the combinations I have used have been conflicts with existing ones.
TK: Does VO read your key combination in the Edit field of the Add Shortcut dialog? It should do that if it accepts the key combination, just navigate around within that dialog before pressing return to check this out. As well, if you navigate around the System Preference, and if there's a conflict that the OS can determine it will let you know in a text field near the Restore Defaults button. I'm not sure about Spark as I don't have it installed.
