Yes, I know. This issue is not to be expected when using the standard setup.
But the impossibility to diagnose an error situation is a bug too.

It's very hard to guess what's so special at your machine to make it fail.
I added the revtex4-1.layout to my private layout files in my user directory
of LyX as you did. But the reconfigure works for me. Therefore I'm unable to
help diagnosing the problem. And I'm unable to create the same configuration
as you're using it. I have Mac OS 10.6.5 and cannot downgrade, I have 
TeXLive-2009
and don't want to update to TeXLive-2010, I tried it with LyX-1.6.9 and don't 
have
the time to install and test it with 1.6.8. None of the mentioned differences
should matter here - but who knows...

So, the missing error output is a problem worth to report it.
