I'm having some issues with P1 transduction in that it isn't working at all.
I've been using a protocol I found online ( http://biology4.wustl.edu/levin/protocols/cloning/PhageTransduction-2008-Brian.pdf) that seems pretty similar to most of the other P1 protocols I've seen around. My titers of phage and bacteria seem right. I'm able to achieve a fairly high titer farming the phage (10^9 or 10^10) on the donor strain and still have an MOI of less than one with the recipient bacteria. Unfortunately, I'm not getting any transductants! My donor and recipient strains are both E. coli B.
