We use the Leica ST5020 here in our cytology lab. We have 3 staining setups. 
One Pap, one Nons/FNA, one rush Nons/FNA. And the Nons/FNA and rush Nons/FNA 
share some dishes.  It would not be possible to add an HE to this machine. And 
as previously mentioned, the GYN and Nons/FNA have to be separate. A lot of 
work to change out dishes and increased risk of error.

feedback on these two stainers and if anyone else is utilizing either stainer 
for both stains.

[Histonet] Coverslip

Hey everyone,

I was wondering if anyone else has seen this occur in their labs and how to 
resolve this matter.  It has to do with the coverslip using the resin tape.  
However more recently, I have been noticing that there is a drying artifact 
that happens right as the xylene drys.  It looks almost like when water freezes 
and gives that frosted look.

We have found this to be a culmination of a couple of things:
Bad lot of tape from vendor
Drying slides before cover slipping
Too much xylene drip during coverslip (dilutes the glue stuff on the tape
allowing for weak adhesive)

We use Sakura cover slipping tape for more consistent quality and adjust
xylene drip accordingly and make sure slides do not sit out very long at
all as xylene from the stainer will evaporate (drying slides) rather
quickly.

[Histonet] Calponin

Hi All,



I was wondering what stain protocols are being used for Calponin to minimize

cross-reactivity with myofibroblasts?

We are using a Dako concentrate antibody. I recently re-optimized the

stain using a 1:100 dilution and changed the pre-treatment from low pH

HIER, to Protease only. This helped a lot with cross-reactivity with 
myofibroblasts

that we were seeing, however we are still seeing some non-specific staining

from time to time. Any information would be greatly appreciated!

[Histonet] possible TB case

We just performed a frozen section on a lymph node that looks like it may have 
TB-  I know we will have to decontaminate the cryostat but what else do I have 
to decontaminate?  Will I have to do anything special to the processor or 
microtome?

Usually these odd results have 2 origins that you should look into:
1- using a different (cheaper and harder) plastic than the originally 
recommended; and
2- a regulation problem in the amount of xylene dropped into the film

[Histonet] IHC Validation:

microarray of different known cases the preferred method?  Does anyone have

[Histonet] Slide Mate Print Quality

In answer to the question about the Slide Mate printing coming off of the 
slides, we had the same issue about a year ago.  At first Thermo though it was 
the ribbon but it turned out the problem was with the slides.  We use the 
Shandon Superfrost Plus (Cat# 6776214).  We contacted Thermo/Shandon and they 
sent us a different lot.  We had to go through about three different lots 
before the problem was resolved.

What is it that you want to know?  I used to make frozen multiple tissue 
control blocks for ER/PR many years ago before these antibodies could be used 
for FFPE tissues.  You could do the same thing with paraffin embedded tissue.  
I've made multi-tissue blocks using a skin biopsy punch tool to obtain small 
amounts of paraffin-embedded tissues to be used in the control block.
