[Histonet] Thermo's Excelsior Tissue Processor Program's

2010-03-30 Thread Akemi Allison-Tacha
Hi All of you in Histoland,

I am working with a facility that recently purchased a Thermo Excelsior Tissue
Processor.  They have had processing problems with several of their specimens
using non-xylene clearing agent.  These issues were with both bx's and larger
tissues.  The program and the non-xylene clearing agent was recommended by the
technical staff at Thermo.

The histology staff have switched back to using xylene verses the non-xylene
clearing agent, and most of the issues have disappeared.

Also, the histotech's were using reagent alcohol, histological grade.  This
grade of alcohol is denatured with methyl alcohol.  The sales representative
was in and informed us that this type of alcohol has damaging effects on
the instrument.  The representative will be coming in to flush out the system
and reprogram the instrument next Monday.

I looked over the current VIP program which is being used, and the program,
timing and temperatures are a little different from most hospital and private
laboratories I have worked with.  We are going to use basically the same
program for the Excelsior, that we use on the VIP.

I would like to know what other Excelsior Tissue Processor users that use
xylene have for their programs.  It would be great to compare the reagents,
programs, timing, and temperatures for Routine overnight runs and for Rapid
Biopsy Runs.  Thank you in advance for your assistance.

[Histonet] Email Spoofed. addresses were highjacked

2009-11-13 Thread Akemi Allison-Tacha
Hi All,

The email below was sent to me in response to the Scam.  I guess someone from
histonet contacted him regarding the scam, which was spammed.  It's interesting
to find out how vulnerable we all are.  Yahoo's customer support has had high
volumes of similar problems.

Akemi,

Boy!  Peter needs an Oxycontin!  Maybe more than
 one.

Don't worry about this.  The correct term is being Email Spoofed.  From my
friends, it happens when some computer hijacker either creates content on a new
or existing web site that you ended up clicking on.  Peter suggests it is
Where you visited, but it could be anywhere, even on a legitimate site.  Some
people I've talked to think Microsoft's web page is loaded with Spoof and
Spyware!

Sometimes, it is a computer worm or virus affecting your computer.  Most of the
time, it is associated with your email
provider.  [Most of my friends with Yahoo email have been Spoofed.]

I would run a up-to-date anti-virus software, and (especially if that doesn't
help) ask for help from Yahoo to quarantine or fix your address.

You probably have it fixed already.  If
not, it will do it again until someone stops it.  It IS only email.  So what???

Phishing also happens over the phone as well. It is not unheard of that
people get phone calls claiming to be from a bank asking for personal
information (why would a bank need your account number... aren't they your
bank and don't they already have this information?). It is generally a very
bad idea to give any personal information out in any venue other than the
physical office of the institution which you do business. If the company
suspects that your account has been compromised, they will disable it and
then ask you to contact them. If you receive such an e-mail, do not click on
any links in it or dial any phone number contained within. Instead, look up
the company's contact information (in a phone book or via a search engine)
and use the publicly available contact information.

2009-10-22 Thread Akemi Allison-Tacha
Hi All,
I think all of you are missing the point of Patti's question.  She stated that
her lab was dinged for having plants in the lab by a CAP inspector.
I had the same thing happen to me years ago.  The inspector stated that plants
attract insects that can contaminate a supposedly clean environment.
Patti has an extremely well organized lab that only had a small phase (1)
deficiency last year.  I think the inspector couldn't find anything, so they
had to come up with this ridiculous infraction.

I don't know Sally, but where I worked many moons ago I had a spider plant that

Hi All.  Just to clarify - the inspection we had was not a CAP inspection..
It was an audit based on the GLP regulations.
Thanks for all the feedback on plants in the

Fw: Re: [Histonet] antibody validation Thanks, but need Non-U also

2009-09-15 Thread Akemi Allison-Tacha

Hi Cynthia,

Thank you, this is exactly what I was looking for.  You are a rare
jewel.  Especially for the size of your facility.  I am impressed!  I wish I
would have received more feed-back from small labs, but that's OK.  I have been
doing some pro-active sleuthing of local labs.

After numerous conversations with my fellow histologists that do IHC, I am
finding out what I have long suspected and have experienced while doing
technical support and consulting.  The routine histology laboratory is
multitasking all of their regular histology duties with the addition of IHC
testing.  Pathologists do not want to lose the IHC revenue to a reference
laboratory, so they bring on-board simple IHC panels, and send out their
esoteric or prognostic markers to a well known reference laboratory.  Most
histologists have been asked to bring on board IHC, without the proper
educational background or tools.    The average histologist has
not had the opportunity to take continuing education classes, or even go on the
histonet.  This may be due to lack of continuing education funds, or lack
of interest; It is amazing that pathologists can go to their continuing
education meetings, but don't support their histologists.  Unfortunately,
these histologists do not understand the importance of standardization or
validation steps.  When something goes bump in the night, they are unaware
how to troubleshoot the problem.  This is unfortunate for the histologist,
the pathologist and most of all, the patient.  One more reason for
pathologists to support continuing education.  Hope to see you all at NSH!

Hi,

I work at 250 bed facility.  For the initial protocol workup we use the
manufacturer recommended protocol and run an IHC TMA control block with 25
different tissues and
tumors.  Once the pathologist has looked and responded I search for 5-10 cases
with expected positive results and then a selection of 5-10 normal or negative
tissues and run the protocol.

Hope this is what you were wanting.

Good Morning and Happy Friday out there in Histo-Land!

I would like your assistance in an issue that I have just become aware of
regarding antibody validation.  I have been requested to bring on-board
approximately (7) new antibodies in the immediate future, and several
antibodies later.

Although, many of you who know me realize I have worked in IHC RD for a well
known IHC company for many years, it has been a while since I have overseen a
clinical IHC lab.  I would like your assistance and input on how you are
validating new antibodies.   Not counting Her2-neu, it has been
my understanding that it is recommended to test samples with varying antigen
expression from 10-15 cases.  Furthermore, you should check for Sensitivity:
expression range of Positive cases, low to high (10-15 cases).  Specificity:
(+) vs expected (-) cases/tissues (10-15 cases) and Precision
(Reproducibility):

It has been my practice that any changes that are made to existing protocols,
such as a new detection kit or antigen retrieval methodology, also requires
re-validation.

I am curious how you approach validation.  Also, do any of you just use a
single known positive control for that antibody, and run it, and if it is
positive, feel that it is satisfactory, and put it on-line for testing.

Thank

RE: [Histonet] Special stains and microwave

2009-08-27 Thread Akemi Allison-Tacha
Hi Rosa,

Excuse me if I am wrong, but I think Bill is referring to using a microwave for
special stains, and you are referring to a Microwave Tissue Processor.

I have used an oven set at a higher temperature (85 -110 degrees C) for
staining the silver portion of the GMS, and it generally only takes between
10-15 minutes.  That is if you place the silver solution in the oven for
approximately 10 minutes prior to use.  I also use the oven set at 60-62
degrees C. for the bouins portion of the Trichrome stain.  Although using a
water bath at 60 degrees C. is the preferred method.

You have more control using a oven than using a microwave.  With a microwave,
results vary with the # of slides placed in the coplin jar, as well as the
placement in the microwave.  There are variations in microwave intensities if
you don't have a carousel.  You also may have boil over of solutions.  Food for
thought.

Bill,
I can highly recommend the EBS processor microwave.  At a base price of 8,900
(last I checked) it is one of the lowest cost processors on the market.  I
don't know if having the flexibility to process in this new microwave may help
justify the price?  I purchased mine from Stat Lab, and the customer service
from the folks at EBS was outstanding; they even had a prob and air agitator
block style holder designed and built for me when I suggested that it should
come with one.  I really like our model, and it offers great flexibility in our
lab.

Greetings Histonetters,

We have finally come to the point where we need to fish or cut bait in
relation to purchasing a laboratory approved microwave and venting it.

While I think it would be cool to have one, I'm wondering if the cost
is justified to do a handful of special stains that could be done
(though more slowly) in a laboratory oven like the one we already have.

Anyone with the time or desire to opine, please chime in.

Those who have purchased such a microwave, I'll gladly take suggestions.

Thanks in advance

[Histonet] Reimbursement for IHC Tests from Medicare/Medicaid

2009-08-20 Thread Akemi Allison-Tacha
Good Morning Histology Managers,

The Chief Medical Director here at my facility proposed a question the other
day regarding reimbursement for IHC testing from Medicare /Medicaid patients.
I asked our billing manager, and she was going to investigate the matter, but I
know my fellow histology managers out there in histoland are pretty up on these
matters, so I am asking once again for your expert knowledge.

The laws have been going back and forth regarding how much we can charge, and I
have been out of the loop for awhile.  Can we charge for more than (2) IHC
tests for these patients?  Let's say you want to do a panel that consists of
(5) IHC tests, can we only get reimbursed for (2) of those tests?  Any
assistance would be greatly appreciated.

Thank you in advance for your information,

Re: [Histonet] (EDTA)Decal Bone marrows

2009-07-13 Thread Akemi Allison-Tacha
Hi Patti,
Years ago, I used a EDTA Decal Solution which was recommended by Dr. Jaffee, a
Hematopathologist at NIH.  This formula was particularly great for BM Core
BX's.  It is much gentler on the tissue, and is less likely to harm the
cellular integrity.  Dr. Todd Barry also prefers the use of EDTA formula's for
BM Bx's.

Below is the formula. or you can purchase from Biocare Medical. You should
decal the specimen for 30 minutes to 1 hour and check for decalcification.  DO
NOT leave for any longer than 1 hour.

10% EDTA (Ethyl-media minetetra-acetic-acid
Stock EDTA 10 ml
DI water   90 ml
pH 4.5-5.0 with Sodium Hydroxide

If you prefer to purchase the IED Solution, Biocare Medical has it available.
They have 2 sizes 140 ml or 500 ml.  cat # IED 1204.
Ion-Exchange Decalcification Unit (IED Unit, Designed for Bone Marrow
Biopsies)

An advanced decalcification system that removes calcium from bone
quickly while leaving superior cellular detail. The IED Unit
incorporates a strong cation ion-exchange resin in a weak acid solution
to remove calcium ions from bone, replacing them with hydrogen ions.
Because the IED Unit does not require strong concentrated acid
solutions, as in traditional decalcification methods, delicate cellular
structures antigenicity remain intact.

[Histonet] Rotary Microtome's and the Meditome

2009-07-13 Thread Akemi Allison-Tacha
Hi everyone in histoland,

I am in the market for 3 microtome's.  I was originally thinking of purchasing
demo or refurbished Microm 355S units or comparable.  I used this unit when I
set-up a TMA lab and was very happy with it.  I am in the process of demo'ing a
Leica RM2255 unit.  The Leica unit has all the bells and whistles the Microm
microtomes has, but it is on a separate device vs Microm's features are created
internally.  I would like your thoughts on this.

Also, I was given some information on a NEW Microtome called the Meditome
Rotary Microtome.  It is made in Germany by Medite and distributed by Micron,
based in San Diego.  It is my understanding that the engineers who made the
Reichert Jung and the Microm microtomes developed this new microtome.  It sells
for under $10,000 and has a 2 year warranty vs the normal 1 year warranty.
Although it doesn't have all the fancy features, it looks like a solid unit.
Has anyone purchased one of these units, and if so, how do you like it?

Thanks,

Good Morning Histoland and Happy Friday!

I need your assistance on a on-going question which has been kicked around
since 2005.  I tried to look this up in the Histonet Archives, but was
unsuccessful.  Perhaps, my key words were incorrect.  Anyway, it was my
understanding that Governor Arnold Schwarzenegger put a mandate on all
laboratory staff, including histologists, that they were required to be
certified by 2011.  It was further my understanding that un-certified histology
staff
were only going to be able to work as histology assistants.  Could I get some
clarification on this matter.

Thanks,
Akemi Allison-Tacha BS, HT/HTL

Fw: Re: AW: [Histonet] AFB Recycling Formalin for Fresh Tissue

2009-05-28 Thread Akemi Allison-Tacha
Hello,
I responded to Gudrun personally, but I thought that the information I am
supplying him below might be a good educational source for other histologists
who are having similar scenario's for AFB contamination.

I would also like to say that I have an extremely eager histology team that
have expressed an interest in learning theory and practice.  Most of my team
are OJT.  They have had several pathologists approach them with these concerns
regarding AFB, and because of their knowledge, or lack of it, couldn't fix the
problem.  We are starting with the basics and I am giving in-services along the
way.

Hi Gudrun,

They are shipping the recycled formalin directly to the outside hospital
surgery to put their fresh tissues into.

As far as the AFB, we have had intermitant (+) AFB on the edges of tissues,
that has been extremely questionable.  We currently are not cutting a negative
(-) positive(+) control on the same water bath as the test sample. That will
be remedied beginning Monday. The water here is also in question.  I will be
sending it out for microbacteria analysis.

We are going through a thorough housekeeping and in-service process.  We are
starting with the tissue processors, which are currently only changed weekly
(not rotated daily), embedding centers, which have to my knowledge, never been
drained, and the hopper cleaned-out.  The water baths are not scrubbed out with
soap and HOT water daily and
covered at night to prevent contamination.  The water bath is not skimmed of
excess tissue debris, each and every time another specimen is placed on it.
The Carbol Fuchsin is not being filtered prior to use.  The working solution is
currently being poured back into the stock bottle.  Need I go on

Just for interest. Your lab takes the recycled formalin to make 4% neutral
buffered formalin (NBF)? Or do they ship the recycled formalin directly in
bottles to the surgery?
AFB = acid fast bacili?

Do you have concerns, that they produce false-positive AFB stains? I cannot
imagine, that a (hopefully) small number of dead bacili on the surface of
tissue will pretend this. An other question: Isn't there a filter in the
recycler to prevent such contamination?

Betreff: [Histonet] Recycling Formalin for Fresh Tissue

Good Morning Histo Land,

I am asking all you out there to give me your input on recycIing formalin.
I realize this has been discussed in the not too distant past, but this may
be a little different situation.  I realize some labs are recycling formalin
to put on their tissue processors, but this is a totally different scenario.

I have been a manager at my lab for only a month.  Much to my surprise, I
just found out yesterday that our lab is recycling formalin and shipping it
out to one of our clients to put their FRESH SPECIMENS into.

I just had a lengthy conversation with Robert Lott the day before yesterday,
regarding pre-analytical protocol's effecting the test results of AFB.  By
the way, we were in full agreement.  This formalin issue was unknown to me
then.  I realize that the pre-analytical, pre-analytical, pre-analytical,
process effects the final out come.  I need to present my case to our
Director of Pathology why this is compromising patient care.

Any and all responses are gladly welcome.

Thanks,
Akemi

Thanks Freida,

What better source to validate the steps I am taking to eliminate the problem
of AFB Contaminate.

I would also like to mention in my haste to compose my e-mail, I may have
stated something incorrectly.  I did not mean that a positive (+) control be
cut at the same time as the test sample and negative (-) control.  I usually
place the test tissue on the same pre-cut positive (+) control slide.

Thanks Rene,

For your input on recycling formalin.  This is good ammunition for my defense
in stopping the use of recycling formalin.  I still would welcome any and all
replies.

Wang reported on the contamination of tissue sections with AFB by the use of
fluorescence microscopy, and if I remember correctly, when he tested the water
in water fountains, he found something like 33% contained AFB.  Non-pathogenic,
but we can't tell that on our stains. The paper is in Am J Clin Pathol 51:71,
1969.  We (Carson, Kingsley, Haberman, and Race) also reported the
contamination in the 1964 issue of the same journal.  We stopped using tap
water for our water baths and began using millipore filtered water that had
already been through a deionizing column and charcoal filter.  We always cut a
negative control from the same days workload as the patient case - uterus in
our case.  You don't need a positive control cut the same day, just a positive
control with only a medium number of organisms.  We
also did not use any tap water in the deparaffinization prior to the carbol
fuchsin.  AFB organisms have also been reported growing in 40 gal formalin
tanks believe it or not and in the old Fisher Paraffin wafers.

We centrifuged tap water in 50-ml tubes, poured off the supernatent, and
refilled and recentrifuged until we could see some sediment in the bottom.  We
took that to Microbioloby and had it cultured to definited prove the presence
of AFB in the tap water.
I would recommend this approach to anyone suspecting tissue contamination.

Usually not, unless the tissues remain too much time in the decalcifyier,
meaning more time than needed to decalcify. For IHC the adverse effect is
greater and because of that, when possible, the decalficication should be done
with EDTA.

Are there negative effects associated with acid decalcification and special

Instead of spitting your venom try to learn

Sort of like you might consider learning correct English? I mean, its
one thing if its not your first language, but despite how many people on
here called you kindly and helpful, I find it perplexing that you feel
compelled to call me a piece of ignorant, and TWICE, no less.

I do not speak French. This list is not in French. Ergo, my awareness
(or lack thereof) of proper French diction has absolutely nothing at all
to do with anything... aside form providing you a vehicle to call me
 names and lash out.

If you have something to say, say it. The simple fact that you do not
like me does not give you- or anyone else- wholesale right to insult me
 or call me names.

You should be ashamed of yourself, Rene. I never expected such
antisocial behavior from you, and frankly, when you behave so
regrettably, I feel no compulsion to learn anything form you!

Hi All,
I guess I was stupid to link on this file.  I just hope aa aa didn't do this
as an act of malicious behavior, and now we have viruses in our computers!  I
usually delete any unknown sender, and fortunately my MAC filters out most

foolishly opened the link to do the same.

Bernie,
I used to use the method below for FS on muscles at Emanual Hospital in
Portland OR. Since we had such wonderful success, we incorporated this method
for all FS.
Years later, I developed the MATSSE (pH3.4) 1-Step Trichrome Stain Kit
(Modified Trichrome Method for Muscle Biopsies Cut on Frozen Sections) for
Biocare Medical.  This kit has long since discontinued by the way...This came
from Biocare's Data Sheet.

NOTE: Muscle tissue should be suitably obtained and
frozen within 30 minutes after excision.  The usual
method of freezing is to first mount a transverse section of the muscle on a
chuck using 10% tragacanth gum or equivalent commercial frozen section mounting
media as the adhesive.  The chuck
with the mounted specimen is then immersed in prechilled isopentane until
frozen.  Isopentane is precooled when a container of the substance is
placed into liquid nitrogen.  At a
temperature of -160° C, the isopentane has a slightly syrupy
consistency.  Care should be
taken to remove the muscle sample when freezing is complete, usually after 25
to 30 seconds.  Too short a
freezing time produces artifacts; prolonged freezing can produce cracking of
the block.

NOTE: If
the sample cannot be sectioned immediately after freezing, it may be wrapped in
foil and placed in a plastic-lidded container along with a small amount of ice
for moisture.  Specimens may be
stored at -70°
C until sectioned.

Does this apply to Liquid Nitrogen as well? I'd hate to have to try to cut
fatty sections in the cryostat without it!

I do mostly cryo, and I've never used liquid nitrogen to chill anything within
the cryostat. What's that all about?

I find that it leaves to many ice crystal artifacts anyway. Not cold enough
fast enough. You might be making it harder on yourself... just set the
temperature of the cryostat for the appropriate temperature of whatever tissue
youre cutting, and you should have no need for a crutch, whether it be
fluoroethane or liquid nitrogen.

Sorry, I too noticed that the text looked weird then I did a cut and paste.  I
keep all the Data Sheets I created in my files, and this was just a portion of
the Notes from the original Data sheet.  I thought it might be helpful because
you stated you had difficulty with cracking.
You did not mention using isopentane.  Some people immerse the tissue straight
into liquid nitrogen, which could cause freezing artifacts.

[Histonet] Sara Sorry to see you go

2009-04-06 Thread Akemi Allison-Tacha
Hi Sara,
I for one will be sad to see your absence.  Perhaps in the future, you will
reconsider and join our group again.  It is extremely unfortunate to see these
grown men acting like FOOLS!

I have been pretty much a voyeur as of late, unless I have something credible
to say or ask, but I feel the urge to state my feelings as a fellow
histologist.  And by the way, there are a number of extremely prominent
histologists and pathologists that have stated they are voyeurs too!  They may
not go on line, but they see your postings!  They have all taken note of this
stupid on-line communication.  I don't think these individuals realize that the
pathology community observes these communications, and this may come back to
bite them in the butt later on down the line!  It lowers the image of these
individuals as professionals, and their names and verbiage are forever archived
in our memories.  Once you ring the bell, you can't un-ring it!
The Histonet Forum was initially created and funded by histology vendors and
concerned histologists to help us as a group.  I maybe wrong, but I thought
it's purpose was to expand our knowledge in this valuable profession.  I have
been utilizing this wonderful tool, since almost the beginning.  It was an eye
opening experience when I started seeing pathologists joining in.  It made me
realize that they too saw the merit in this wonderful tool for sharing our
knowledge and experiences.

I have been involved in histology since taking classes in histomicrotechnic at
Tokyo University in 1968.  The older I get, the more I realize how little I
know.  Especially with the use of molecular biology, and more and more esoteric
testing.  We are experiencing a metamorphosis in our field, and forums such as
this, are extremely valuable!
Those of us who use this tool, do so, because we care about giving the best
possible patient care.  At least I use to think so This is a wonderful
tool for histologists, researchers, and concerned pathologists, to come
together for the sole purpose to learn and grow (then there's the Friday good
hearted release of steam)  The Histonet is especially important for those
individuals who are geographically isolated, and do not have the means to
attend continuing education classes.
It is my belief that the Histonet was never intended to be used as a tool as
these individuals have so irresponsibly done.  It is childish and
unprofessional.  This sort of use just shows how unthinking and selfish these
individuals are.  I am ashamed that the pathology community sees this sort of
communication between histologists.  It demeans us as a whole.  I don't think
these men realize what fallout may occur.  We may have this taken away from us
because they have not used their good judgement, common sense, and restraint!
I realize that in the past, I too may have spoken a little out of turn, but
this thread has become an embarrassment.
I have recently accepted a employment opportunity to go back to managing a
histology lab, and will ask my fellow histologists for their assistance.
I truly hope there will be a Histonet available when I need you!
Let's hope we can learn from this experience and move on.

[Histonet] Roche/Genetech Today Buy-out and Lay-offs

2009-03-13 Thread Akemi Allison-Tacha

FYI... This is from Todays Fierce Pharma Report:
Today's Top Stories

1. Roche to cut jobs in California cowboys deal
By Tracy Staton Comment | Forward
Deal dissection of the day, No. 1: Now that Genentech has finally conceded to
Roche's buyout plans, the postgame analysis begins. As we all know after months
of following the possibility of a deal, preserving innovation at Genentech is
job one. Next on the priority list comes cost-cutting. But neither of those
goals will kick in if shareholders don't tender, so we're hearing plenty about
their opinions. Here's a sampling of the predictions, prognositications, and
pushy advice:

Roche plans to shed more than 1,500 jobs in New Jersey and relocate major
operations to California as part of the Genentech takeover. Some 1,600
researchers and scientists will stay behind in Nutley, NJ. But the company is
redoing its campus master plan to figure out what to do with all the vacant
buildings that are left. And who's first in line for the job cuts? None other
than sales and marketing, the Star-Ledger says.

After years of partnership, Roche and Genentech are already close. Merging
their drug portolios should be a snap, New York Times DealBook says. But their
cultures are almost diametrically opposed: Roche is Swiss;
trains-running-on-time, precision-watchmaking Swiss. Genentech, as one Silicon
Valley venture capitalist put it, are a bunch of entrepreneurial California
cowboys. Get the image? Then you see the problem.

Roche execs may be guzzling celebratory champagne (somehow we can't imagine it,
but it's possible). But along with its buyout of Genentech's promising pipeline
come worries about payer reimbursements. President Obama's push for
comparative-effectiveness research could leave some Genentech drugs struggling,
BusinessWeek reports. Some previous head-to-head studies have shown the
benefits of expensive cancer biologics are small; if that trend continues, the
bottom line could suffer.

several divisions that produce products that compete directly with some of
TBS' industry leading reagents and consumables, and, has more recently been
acquired by Thermo, the parent company of Shandon.

 Hi Doug,

Thanks for the clarification. You are absolutely correct!

Have a great weekend fellow histonet subscribers! As for
me, I'm packing and heading back to the SF Bay Area. I
have accepted a position as a Histology Manager for a
private reference lab. My heart has always been in the lab
 and patient care.

 Wish me luck,
 Akemi

You certainly do need a program to keep track of who
 is who.
  Didn't Fisher
Scientific purchase Apogent and the Thermo purchased
 Fisher
  the next year to
form Thermo Fisher Scientific? If so then Thermo
 should be
  the parent
  company.

  Happy Friday the 13th Histonet!

Forewarning, this is a little long. There seems to be
 a
  lot of discussion as
of late, to all the buy outs, and who owns who. I
 have
  been following this
trend for several years with great interest. It is
 amazing
  how pricing,
customer support and service, varies from one company
 to
  another, even though
they are owned by the same parent company. I can
 certainly
  understand and
relate to the frustration and anger of the end-user.
 There
  is so much
disconnection within the companies! We need a score
 card
  to keep up with all
the changes! I maybe wrong, but I don't think the
  sales reps even know the
ownership tree. I put a tree at the end of this
  correspondence that I think
  is correct.

I recently had a conservation with a HR rep from
  Thermo/Fisher, and she
didn't even know who Peter Scheu was. Pretty
 telling
  of the knowledge out
there. HR should at least know who is at the top of
 the
  chain!

Quote: Peter Scheu has been Group President, Clinical
  Diagnostics of Apogent
Technologies Inc. since September 2000. Mr. Scheu
 served as
  the President of
  Richard-Allan Scientific Company
  (Richard-Allan) from 1997 to 2000 and as
Executive Vice President of Richard-Allan from 1995 to
  1997.

Let's dispell the confusion and please correct me
 if I
  am wrong.

  The ownership trees:
  Parent Company: Apogent Technologies Inc
  Thermo/Fisher
  Richard Allan
  Microm
  Lab Vision
  Neomarkers Antibodies

  Parent Company: Danaher
  Leica Microsystems
  Vision Biosystems (Bond)
  Novacastra
  NEW-Suripath
  NEW-McCormick

[Histonet] back to the lab in SF

2009-03-13 Thread Akemi Allison-Tacha

Hi All,

I have been receiving numerous inquires as to why I have left PhenoPath. I
appreciate your inquires and congratulations. I am going to give a blanket
reply, since I have time constraints.

PhenoPath is a great company and I have the highest regard for Dr's. Allen
Gown, Steve Kussick, and the rest of the staff. Sometimes there is a bigger
plan in the cards for us, and my talents are more suited for the laboratory and
direct patient care.

I was offered a wonderful job in Los Gatos, CA, which is 30 minutes from Santa
Cruz, and an hour from Carmel and the wine country. I love the Bay Area. I
lived there for 8 years while working for Biocare as the Director of Histology,
and I have a lot of friends there, plus weather is wonderful, and it's like
going home!

Regards,
Akemi

2008-12-20 Thread Akemi Allison-Tacha
Hi Histonet Subscriber's,

The following forwarded e-mail was from Chuck Churukian. I spoke with his wife
Irene last evening and she wished to have this Holiday wish forwarded to the

Thank you for your Greeting that included every religion,
 every circumstance, situation,
sex orientation, etc. etc. It was very humorous.
Here's our greeting As simple as
 we are...

Names We Hold Dear

 by Vicki J. Kuyper (paraphrased)

Every name is a touchstone that leads to a place and time,
Where God has used another’s heart to reach out and
 touch mine. (ours)
It may have happened years ago or even yesterday,
But every person on our list has changed our lives some
 way.

Through simple conversation, a warm hug or a shared meal,
Every person on our list has helped us grow or in many
 ways- heal,
Or laugh or love or learn or smile…..the blessings never
 end
As God allows our paths to cross as family and/or friends.

So please know that this greeting is more than a Christmas
 wish.
It’s a “thank you” to God for putting on our
 list---
Each and everyone whose name we have come to hold dear….
Those who have shown us Christmas love – years passed,
 even this year.
Have a wonderful holiday. Get some rest and
 relaxation
Our wish and prayer for you:
A Happy Healthy Prosperous New Year
Chuck and Irene

Re: [Histonet] Re: Fungal stains political views

2008-11-05 Thread Akemi Allison-Tacha
Hi All,

I totally agree with Robert on his information regarding periodic acid and
chromic acid. I have done extensive research on this subject when I developed
my special stain kits for Biocare Medical. I did incorporate 10% chromic acid
for the GMS stain, and 1% periodic acid for the PAS stain.

As far as this forum for political views, in frustration, I deleted everything
with these headings. There could of been some very interesting feed-back, but
you have used this source totally inappropriately! I believe in this countries
beliefs for freedom of speech, but use it wisely!! The histonet is NOT THE
PLACE!

Colin Weaver asks about staining of various fungal species
 by the
 standard fungal stains.

The standard fungal stains have two basic steps:

 1. Periodic acid or chromic acid splits bonds between
 vicinal diol
groups (two carbons with OHs on adjacent carbon
 atoms) and oxidizes
the free ends to aldehyde groups (-CHO).

 2. The aldehyde groups oxidize Schiff's reagent or
 methenamine silver
 to produce a visible deposit.

Periodic acid is not a sufficiently strong oxidant to
 demonstrate some
fungal cell walls. In human pathology, Histoplasma is most
 likely to
stain weakly or not at all. You need chromic acid, most
 usually
 followed by methenamine silver.

Unfortunately, many so-called GMS kits silently substitute
 periodic
acid for the more hazardous chromic acid. Freida Carson
 reviewed this
top in J Histotechnol several years ago - I can find the
 reference if
anyone needs it (I think I've posted it on Histonet
 before, so it's
probably in the archives.) So your first step is to see if
 you're
 actually using chromic acid.

PLEASE get the politics off this list - the repetitious
 garbage is
making the list hard to read, particularly in digest form.
 Linda
Margraf, if it doesn't stop, I suggest moderating the
 list for a few
 days, though not permanently.
