Melt down old blocks, have them embed and cut them. Evaluate their time and 
technique. You could also do a working interview where a tech shows them around 
but has them cut and embed a few current tissues while watching. Thanks Anne

[Histonet] Small processors

We are looking forward to ordering a new or used processor. We have the Sakura 
VIP5 however, I was wondering if there were smaller processors out there. We 
only do a max of about 60 skin specimens and the VIP is a bit large for that 
quantity. I would say a max of 100 specimens for future growth. Any thoughts.

We have 3 new leicas and we constantly have to adjust them. They creep forward 
so much that I just automatically know to roll it slightly back after every 4 
specimens or so. The new leica designs are just bad and customer service is 
even worse. We have to use the plate as we do Mohs specimens and the sections 
are just better with the glass plate. I want to look for a different machine 
next time. We did test the Avantik and it cut nice but the glass plate was on 
the front and got in the way. I wish they would talk to Mohs techs before they 
design these. Oh well. No tricks just constant adjusting. Thanks Anne

Hi everyone. Does anyone regularly use the anti-roll plate on their cryostat? 
Does it work well on the Leicas and Microms, or do you have to figit with it a 
lot? Does anyone have any tricks they can share to get it to work all the time? 
 I found that a little spritz of freeze spray on the plate and the bottom of 
the roll glass helps, but it is still seems a lot easier for us to use a brush 
or a teasing needle.

[Histonet] Stain line order

When doing frozen section staining for Mohs we have DI water and don't need a 
clearing agent. I am helping set up someone who is using tap water and needs to 
clear it. They mentioned that they used to clear first, rinse, then use bluing. 
I thought it was the opposite after heme. Rinse, bluing, rinse, clearing. I 
don't remember which is first. Actually do you need a bluing if using tap water 
since that blues better then DI water? Thanks for your input, it's been 10

[Histonet] Cryostats for Mohs specimens

We are shopping for a new cryostat; has anyone used the Tanner Scientific (r) 
TN50 cryostat? Can you let me know if you like it. We are unhappy with the new 
Leica's. We are also looking at the Avantik ones as well. Any input is helpful; 
this is for Mohs specimens so they need to cut well and be able to get a lot of 
tissue on each slide, sometimes the components of the machine get in the way of

[Histonet] Frozen tissue

I'm testing a faster Mart 1 for melanoma on Mohs specimens. Its been a while 
since I've done any frozen tissue IHC stains and I can't remember the fixation 
to keep it viable for testing the next day, or same day but later on. Will it 
hold up if fixed in acetone and dried or do I need to refrigerate or freeze?

I have a back-up processor we keep filled for a back-up and if we have an extra 
run. However with lower volumes lately we aren't using it. I was just wondering 
about the paraffin in particularly. Does anyone know if you are not using it 
how long it can stay there (its heated) before changing out completely? Does it

[Histonet] Formaldehyde test kit question

We switched formalin neutralizing solution and I bought the Quantofix 
formaldehyde test kit to make sure it's working, however it does not mention 
what color is acceptable for neutralization. The options are 0-10-20- up to 200 
MG/L. should it be "0" for acceptable dumping? Also the instructions say to add 
10 drops of a sodium hydroxide solution to the solution being tested, wouldn't 
that change the Ph and make the results not as accurate. This kit is expensive 
if regular pH strips are better, let me know that too. Thanks for your help.

[Histonet] Xylene and Formalin monitoring

[Histonet] Grossing large ellipse melanomas

We are starting to keep our melanomas in-house now that we have a better 
processor, however I could use some tips or tricks on cutting large fatty skin 
ellipse specimens. My problem I think, is too small a knife, the specimens are 
not cutting even and the fat is getting in the way. So any suggestions or links 
to grossing techniques would be appreciated. Also, any ideas on better cutting

[Histonet] Slide disposal

When disposing of slides after the 10 period is up, what is everyone doing to 
protect patient confidentiality? In the old days we would just put packing tape 
around the box and toss. With patient names on the slides and sometimes 
specimen location on the new labels I'm wondering how to dispose of them

If you don't already have it you may want to offer a shift differential for 
very early or late shifts and weekends.

We had an inspection from the fire department recently and they said we need to 
have the exhaust from the fume hood checked annually.  Does anyone know who 
does that?  The people that do our P.M. on our machines say they don't, so who

You clearly don't know your histo history. The reason we know that H pylori 
exists is because a Scientist, Dr. Barry Marshall wanted to prove bacteria 
caused ulcers and not stress.  No one believed him.  So he took the organisms 
from a patient, mixed it in a broth and drank it. He then biopsied himself and 
treated it. There's a non-uniform method that saved a lot of suffering.  Bravo

There is a grandfather clause if you were a histotech by april 24.1995.  I 
asked ClIA recently when they came to inspect me, as I fell into this category 
but didn't know if you had to have the licence by then or just have been 
performing high complexity test at that time (my licence was later than that).  
The CLIA person couldn't decipher their own rules and was going to ask her 
supervisor.  I never heard back.  As for IHC any HT in our state can do it and 
CAP never brought it up when I worked at the hospital.

First make sure your block is cold.  Second, some models of microtome's and 
blades don't cut well without dulling the blade a bit.(newer models don't seem 
to need it.)  Lastly some processors don’t fix as well as others you may need 
to face the blocks in and soak them.  Again some places soak in ice water 
alone, some use fabric softener on the ice and some use ammonia water. (that 
one is stinky). Some places do soaking at all.  I now have a really old 
microtome and have to blow or breath on the paraffin lightly as I'm cutting, an 
old technique but effective.  Sorry its going to be a lot of trial and error.  
Maybe you can call the previous tech on how they did it there.  Anne

that does not have someone dedicated to cutting. I already miss her.

I have no problems getting ribbons of 10-30 sections long but the pieces
are half the size of the original block. I am guessing they are wrinkling.

[Histonet] RE: embedding

My first job the molds were kept room temp or cool and I didn't have to scrape 
as much. My 2nd job they were kept hot and had to scrape all the time.  Have 
fun with it and test some cold, some room temp, some hot see if there is a 
difference. The problem with too cool molds is it takes a bit longer to embed 
cause the bottom takes longer to warm up so it's a trade off for time. Fun 
experiment though!  Anne

I mark shaves and punches with .5% Toludine blue it won't color the skin but 
will color the rest.  And best of all it doesn't show up in any staining its 
purely for identifying your tissue for embedding purposes.  Anne

On fatty tissue, especially cheeks. After we mount on a slide we spray the 
tissue with liquid nitrogen and let sit to freeze before embedding for a couple 
of minutes.  Than after embedding spray the chuck again with liquid nitrogen.  
On really fatty stuff you may have to freeze before cutting each time. Time 
consuming yes.  But I find freezing and let sitting I don't fight with it as 
much when cutting.  Anne

Hello,
   My name is Sylvia Shockey I work in a histology lab.
One of our microtomes is not working properly and would appreciate some
advice on purchasing a new one or maybe refurbished.  I have had some
quotes on a Tanner scientific manual rotary microtome, titan 5000 .
Thermo Shandon Finesse and a Thermo Shandon Microm HM325. Mr. Grimm with
Thermo fisher has new and used on both of these models. Help me please I
don't want to buy a clunker.  Thanks for your time.

We got some plexiglass dividers at the Container Store if you have one
near or check online.  Can't remember what they were called but they had
different sized square compartments that fit various venders antibodies.

Does anyone have any good ideas for keeping the little vials of
concentrated antibodies neat and organized in the fridge?  Right now we
keep ours in empty pipette tip boxes (without tip holders) but they're
always falling over and mixed up.  We use several different vendors for
our concentrated antibodies, so our vials are all different sizes.

For the most part I like this system.  A few things though.  Their HE
racks constantly break so make sure in your contract you get free ones
to replace them.  Also if you have to remove a coverslip it is a pain.
You have to heat the slide for a while so see if they will include a
heater and you will need it.  Every once and a while the coverslips get
a bad lot and you get bubbles.  Also the color of your cassettes can
make a big difference on if the barcode reader can pick up the code so
ask them about it. I don't work there any more so maybe they have fixed

building a new lab...they want to sell us their vantage tracking system
and HE stainer.  Our Rep is coming on Fri to talk to me, our lab
director and CEO about these products.  Anyone using these?  I'd like
some feedback so I can be prepared with questions on Fri for our
meeting.  Thanks!

easy access,  Inspectors will ask employees if they know where it and

We had same problem at my old job.  I don't think they have come up with a 
separate IHc so our doctor would order an alcian yellow with ihc on children 
under age of 12 I think but we gave this up cause it's so rare. If you look up 
on Google under h. Heilmannii,  I believe its spelled, it has a lot of papers 
written on it that might help.  Anne

If anyone wants to cut down on cutting injuries.  Teach the new techs to
use two forceps instead of your fingers.  I'm too old to change but I
know a tech who cuts this way and has never had an injury.  It works
really well on automated microtomes.  Anne

I am curious about the rate of microtome cuts that students receive in
Histotechnology schools. We have a rather large program and take in
approximately 60 (or more)  students a year. Our Histotechnology
semester is around 20 weeks long. During that time our students cut
approximately 300 - 400 blocks per student. We typically have
approximately 10 minor cuts per semester. How does this compare to other
