Hi Cheryl,

I have heard of people using cucumber and processing it, but I have not
actually tried it out myself! One of my R friends recommended it to me
when trying to DIY some tumor microarrays. You might be able to try it out
and see if it stains well - I have never actually seen it in person myself!

[Histonet] Leica Slide Printer

Hi all,

I am looking to see if anyone here uses the Leica HistoCore PERMA S Slide
printer/s in their lab? The integration with the Bond III IHC stainer makes
them highly appealing to my team since it will allow us to error proof the
IHC staining process. However, my finance/business folks are worried since
they are guaranteed for 1 year that they will break down after that time
and then we will play the replacement rodeo on our dime.

[Histonet] Dako PD-L1 questions

Hi all,

I am looking to connect with people that stain PD-L1s on either of the Dako
Autostainers. My company has been asked by a research/pharma partner if we
would switch our IHC staining system to this brand. We currently use the
Leica Bond IIIs. What I am curious about is how long a run takes, how
reliable you find the instrumentation to be, and how user friendly it is.
Additionally, does it require a lot of maintenance?

Hi Chakib,

I am not sure what kind of budget you have for control tissues, I work in
the private sector unaffiliated with any one medical center so we routinely
have to buy tissue for control blocks and validations of new
antibodies/assays. We use two different suppliers - RS Diagnostics
 and Cooperative Human Tissue Network (CHTN
) They can be pricey, but that may be due to our
business structure and lack of signed agreements to commit to a specific
quantity of samples over time. I hope this helps!

Hi Blanca,

I think practical skills tests are a great idea. BUT I also think you need
to make sure that having someone that isn't an employee doing tasks in the
lab is a responsibility your lab would willingly take on if something were
to happen. If the candidate cut themselves and was litigious they could go
after your lab. I have not heard of it happening, but I know in this day
and age it can be a risk. Otherwise I try to ask probing questions -
usually things that cannot be answered with Yes or No. Also, I try to ask
about things you really want to know - how do they manage stress, do they
have troubleshooting experience, if so can they explain the problem and how
they solved it. Are they intuitive enough, that looking back on a
troubleshooting or stressful event can they identify what they would have
done differently? I want to make sure that the people I hire can manage
stress, and think through an issue. I also try to ask them what they feel
makes a good tech, and how they think their coworkers would describe them.
It gets them thinking about the team, not just how they see themselves.

[Histonet] Tissue scrolling

I am curious if any of you perform scrolling of paraffin embedded tissue
for molecular studies? If so, what precautions do you take to minimize
contamination? I am asking specifically about at the cutting station as
that is my area of expertise. But we are looking to pilot this for samples

Hi Jennifer -

It was my understanding that the recall was particularly related to the
flammability of the freeze spray. If you can find a spray with out a
flammability hazard you can keep using the spray. I don't have a lot of
frozen sections in my current job. But in my last job, and my experience in
hospitals to get rapid results for patients still on the table sprays are
used. In cases of TB they are discouraged.

I agree with what Toysha said, and also don't take your more experienced
techs for granted! There is great value in techs that have been at this for
a long time. They may not seem tech savvy, or fast to you now, but some of
them have been at this longer than us younger techs have been alive! Some
of the greatest cutting troubleshooting tips I have came as the result of
the knowledge of these veterans! You truly do not realize how helpful they
are/have been until they are no longer your neighbor to ask questions of!

[Histonet] RUO antibody

Hi All,

I am looking to see if anyone has had any experience with the antibody AXL.
We are trying to work it up on our Roche Discovery Ultra and it does not
seem like it stains both nuclear and cytoplasic cell features. We only seem
to see cytoplasmic staining, and in general most tumors are showing
negative results.

I currently am using the Ultraview kit for optimization, our pathologist
likes CC2 for 44 min, Ab. Inc for 40 min using a 1:100 dilution.

Anyone have any suggestions that might help? If you want more detailed info

[Histonet] LDT question

Hi All,

I have a validation question - we are looking into buying an RUO rated IHC
stainer for a future multiplexing project. Roche's Discovery Ultra to be
specific.

I have a pathologist that is convinced that we can run our clinical
antibodies on it as an LDT and still bill for the work. The material I have
read up to this point regarding that instrument explicitly states that no
slides can be run for diagnostic purposes. The instrument has software that
allows you to use it to mimic the IVD instruments' software capabilities so
you can validate a protocol on it, then move it right over to the Benchmark
so I can see why the idea that it can run our IVD items seems plausable.
However what I need to know is which is correct?

I do not want to do something that I am uncomfortable doing without
checking with others in my field first.

[Histonet] Molecular Testing

Hi All,

When cutting for NGS testing what additional measures do you take to
minimize contamination? We are a pretty clean outfit here, but still, have
some trace sample carryover occurring in cases we cut in-house. We already
use a fresh blade with every case, use tools like forceps instead of
fingers to transfer sections from our blade to our bath, and wipe down the
blade holder and water bath surface between every case.

Our NGS team is finding very small - like flake sized bits - of
contamination in samples, we do not know for sure that it is from cutting.
However, they want all departments to troubleshoot. Any suggestions would
be a great help!

[Histonet] Whole Slide Imaging

[Histonet] Suggested Requirements for new uncertified HTs

Hi All,

I work at a Start-Up and I was our first hire. I have been practicing since
2011 when I got my certification. However, we are looking to expand, what
would you suggest our baseline technical competencies be for a position
that focuses exclusively on cutting and IHC. In an interview it is hard to
access what a person is capable of, would it be acceptable for me to have
applicants performs a cutting time test?

I am open to hiring new techs, but I want to make sure we are not hamstrung
by a great interviewer with a lacking technical skill set.

[Histonet] HPV DNA Probe question

 I am curious what people are using for HPV probes. We recently stumbled
upon the fact that our current probe choice is RUO - to avoid a sticky
validation and LDT process I would like to replace that with an ASR or IVD
ideally. I cannot seem to find one, however. Additionally, our Pathologists
prefer HR and LR be run on separate slides instead of as a cocktail.

Anyone have any thoughts?
