We have found the same thing.

You get a moderate staining (half of what you see with most B cell lymphomas) 
with clone 1EW (as supplied with the Leica Bond rtu).
You will get similar results with clone 24 (38%:  Adams, et al (2009). 
Clinical, phenotypic and genetic similarities and disparities between 
post-transplant and classical Hodgkin lymphomas with respect to therapeutic 
targets. Expert opinion on therapeutic targets, 13(10), 1137-1145.)

We have found that using the Rabbit monoclonal antibody to Pax-5 (Clone SP34), 
it seems that, though staining a similar proportion of B cell lymphomas, 
staining is absent from the Reed-Sternberg cells of Hodgin lymphoma.

Our assessment is in its early stages so with further cases, it might be 
clearer.

It seems that not all Pax-5 antibodies react the same

My pathologists are not happy with PAX-5 on the Ventana Ultra; it will 
occasionally show cytoplasmic staining of the Reed-Sternberg cells (supposed to 
be a nuclear stain).  We use Ventana’s clone SP34. I have been trying to 
optimize with the Optiview kit but we are getting so much background staining 
and only sporadic staining of the R-S cells.  I tried a different clone, Cell 
Marque EP156 which was really clean but would not stain the R-S cells at all, 
no matter what we did.   My Ventana technical specialist is stumped as to why 
we’re having so much trouble.  Does anyone else have the same problem or have 
any suggestions?

[Histonet] PAX-5 staining problems

Good Morning to all:  I am having staining problems with my PAX-5 predilute 
from CellMarque.  I was wondering if anyone else has experienced a dramatic 
dropoff in staining signal with this antibody after a few months.  I have now 
had this happen to me twice in the last 3 months.  It stains very nicely when 
the vial is first opened and seems to hold staining intensity for about 3 
months and then drops off dramatically even though the expiration date is 2 
years out.  I am using it on the Leica Bond.  I have been in contact with 
CellMarque about this problem but they tell me they have not had any complaints 
from any other labs.  I use many antibodies from CellMarque and have only seen 
this problem occur with one other antibody (ALK-1) about a year ago.  If you 
are experiencing this problem in your lab please contact Liz Beitman at 
CellMarque (lbeit...@cellmarque.com) so that 
they can have all available information to further research this problem.

Just about any tissue should work.   It stains for Hematopoietic 
stem/progenitor cells, bone marrow stromal cells, endothelial cells, embryonic 
fibroblasts
We use a sausage style control here with about 30 different types of tissue.

Joel,
A section of skin will be a fine CD34 control. All collagenous connective 
tissue, including the dermis,  are rich in CD34+ dendritic interstitial 
fibroblasts as well as CD34 positive endothelium in the resident vessels. 
Actually, in my lab we use a section of fallopian tube from tubal ligation, 
they are loaded with the fibroblasts and vessels.

We use Cell Marque's (via VMS) clone 24.  We have to throw the entire
arsenal at our disposal at these stains but they're acceptable.
