You have to configure/check the SubTone.sqf file and then when this is
setup
you can use the T0/T1/T2 option within SatPC32. Default behaviour is T0
no tone
T1 is normal TX behaviour and T2 arm the SO-50 timer.
Here some information from the SatPC32 Manual:
Hello Les,
with the settings that come with the SatPC32 12.8b setup file you can switch
on the 67.0 Hz and the 74.4 Hz sub tones
of the IC-910H. While tracking SO-50 click the SatPC32 T control (in the
uppler left part of the SatPC32 main window) to T1. That will switch on
the 67.0 tone.

Thank your the advice on how to improve my satellite operation. Much
appreciated, and exactly what I was hoping for when I posted the questions
to the AMSAT BB. Tried it last night on a pass with mixed results. But
I think I understand the principle now.

Sometimes the offset in the transponder changes slightly. This is especially
true with VO-52 and AO-7. On these sats, I use the RIT to touch up and make
sure I can hear my self plainly at the beginning of a pass. This is a little
easier than the offset feature in the software.

Here is the procedure I've used with SatPC32 and my Icom IC-821 for
Doppler correction when operating SSB. It should work with any rig that
is CAT controlled. (This is a corrected version of something I wrote a
