I don't think it is a "standard" procedure. However, our pathologist will
frequently ask us to keep intervening levels for possible IHC on certain
cases that are small or where the tumor is about to be exhausted. We are a
dermatopathology lab. When it is requested, we cut the whole case

Cap requires a negative with each block of patient tissue being stained for
immunos, However they also state that if more than one block from the same
patient is being tested that only one of the blocks is
CAP guidelines clearly state that you must run a negative control for each

antibody, you may validate your procedure to run only one positive control.

I have seen this problem many times over the years. Nearly 100% of the time
it is due to the level of the alcohol after the eosin being too low.
Whether you water rinse or go straight into a 95% etoh, the level of your
first alcohol after eosin MUST be covering your entire slide. If not,
and could you give me any advice?
All help would be greatly appreciated. Can I make a 50/50 mixture of the
RTU antibodies and use my longest protocol? We are using the Ventana Ultra.
