I don't know a method to obtain selective red coloration of Gram-negative
organisms, with yellow for both Gram-positives and "background", and I cannot
find one by looking in various books. It may not be possible because
Gram-positivity relies on selective retention of an immobilized dye.
I do a gram stain but also not very often. I have used a PolyScientific kit and
it has a pretty good shelf life but before that I was just making up the stains
and solutions in small quantities. They aren't hard to make and I inherited a
wealth of stains and chemicals here that it seems a shame
We have had great success with the Twort gram stain.

We use a Twort stain with my students. They have been doing the gram

the entire procedure.

Stacy,
You can buy a complete Brown and Brenn staining kit from Newcomer Supply
which is very good, and not change your method. The picric acid/acetone
mixture is included and you wouldn't have to store stock picric acid nor
ether in the lab but just this small amount of Picric acid/acetone
Thank you!!

We uses 1% aqueous neutral red as red counterstain
with very good results.
Do not forget rinse slides at 10-15 secs in DW
with 1-2 drop of glacial acetic acid before neutral
red and after this, because this dye have red color
at pH 6.

I always used actual tissues with gram +/- bacteria because it is not the same
(time wise) to stain a histogel than a piece of tissue. Appendix is a good
control.

Thank you for all replies.
Just a quick update: I have carbol-gentiana-violet, of course, I just forgot to
mention it.
Please tell me how to decolorize the fuchsin without affecting the violet, that
would help me very much in this moment. Thank you!
I'll post my results and my method of
