No, you definitely want to install it before the OS.

I've only recently used it with Linux as a the host but the USB passthrough
should be seamless and toggleable on poweron of the VM -- and as long as
Windows has drivers for it then it should see it. The extensions enable USB
2 and 3, however, so unless you have a really old USB device then it
probably won't be seen unless it has backwards compatibility with USB 1.
