[Histonet] John Caggiano’s passing
Dear HistoPeeps: I have some sad news to share with the histology/pathology community. Joe Caggiano informed me Monday that his brother John passed away this last Sunday night. John Caggiano, founder, President and CEO of Poly Scientific R & D Corp, died quietly in his sleep after a long illness. John and Poly Scientific were early and longtime supporters of NSH, (National Society for Histotechnologists). He was always supportive, generous, and it was a pleasure to spend time with him. John was a great friend and he will be greatly missed. RIP John. Re: [Histonet] Faded H tissue section
Hi Tony: I actually gave that tip to Lee back in 1979 when he came to OHSU to give a seminar to the Oregon Histology Society. I found out that when I was doing GMA and the pathologists didn’t like Gills Hematoxylin. They loved my PAS on GMA so I tried using 1% Periodic Acid before using Harris Hematoxylin for H’s on GMA and it turned out beautifully. Guess he shared my tip with the rest of our society. He and I became closer se friends and we shared several tips, including his Movat’s tips which he didn’t publish, but I shared them in Frieda’s 4th Ed. [Histonet] Validation of new G2 Coverslipper
Good Morning Histo Peeps: We just purchased a new Sakura G2 Coverslipper for our IHC Lab. The engineer will be coming Monday to make sure all adjustments are in alignment and fine tune anything which might have gotten out of alignment due to shipping. I have never validated a new Coverslipper in any of the labs I have managed. I searched the internet and was not able to find any information or instructions pertaining to Coverslipper Validation. I am of course going to run several test slides prior to running any actual cases on it. I will instruct my staff to date and provide test information on the slides to accompany the paperwork Sakura provides for the set-up of the equipment, but I am not sure that will be sufficient. Since we our a research facility, my IHC Director does not feel we need to validate our new Coverslipper to comply to GLP. I just want to make sure for my own assurance. I would greatly appreciate any guidelines for validation you may want to share with me. [Histonet] GLP lot# documentation
Hello Histo Land: I am the new IHC manager for a research lab which works primarily with animal tissue and we are moving towards GLP compliance for our department. We have an extremely large client base which consists of Big Pharma, private research companies, universities, etc… We are in the process of updating our SOW and SOP forms, Stain line maintenance forms, solution stainer change logs, etc… I have a question regarding documenting lot #’s for our hematoxylin counterstain protocol for IHC's. Do any of you who are GLP compliant document lot numbers for your alcohols and xylenes? Our director and QA officer would like to incorporate documenting lot numbers for alcohols and xylenes. Our histology department who is GLP certified only documents their lot numbers for Hematoxylin, Eosin, Bluing solution and High Def solutions for their H stain line. They currently do not document lot numbers for alcohols and xylenes. I have primarily worked for Clinical pathology labs, who did not document lot numbers for alcohols or xylenes. Please let me know if you do. [Histonet] Bx Processing run
Good morning Histopeeps: I am reaching out to you this morning to ask for your processing protocols for your biopsies and routines. Our department has two 25 year old VIP Tissue processors. Prior to my arrival, the department was only using one processor at a time and running the biopsies and routine tissues together. They were also only rotating solutions once a week even though there were close to 300 cassettes a day. With that being said, we are now using both tissue processors and separating biopsies from routines and we rotate all solutions daily. The specimens are put on the machines at 5:00 PM. This is not an option. Our biopsies finish at 5:00 AM and the routines finish at 6:30 AM. I would like to have our biopsies finish by 3:00 AM and the routines finish by 5:00 AM. Naturally, I do not want to compromise the quality of our tissue samples. I would appreciate you sharing your processing protocols with me which would include solutions and time in each station. [Histonet] Tissue disposal
Good morning Histopeeps! After reviewing our Policy Procedure Manual, our manager is questioning the bags we are using for disposing our tissues after diagnosis is complete. We are separating the tissue from the formalin by straining the excess formalin and neutralizing it. We then put the tissue into “Large Biohazard Bags” and the EVS staff picks it up and disposes it. My manager is wondering if the “Biohazard Bags" are appropriate. Should the bags state “Tissue” on the them? Are there any special disposal bags available? While working for a GI Lab in Monterey, CA we put all our bottle and tissue in "Biohazard Bags" and a Hazardous Waste company picked up the waste products from our endoscopy department along with ours. [Histonet] Histo techs assisting on BM Bx's
Good Morning Histo Peeps: Back from a great trip to NSH NOLA and hit the ground running yesterday. I am faced with a department coverage issue which impacts our TAT. I am curious how many facilities have their histologists assist with BM Bx’s. I have never in my 50 years as a histologist assisted with BM Bx’s. That responsibility was under the hematology department responsibilities. We have 4 FT Histo tech’s and 1 PNR in our department. Yesterday we had 2 histologists pulled away from bench duties to assist with BM Bx’s, and then another histo tech was called to go to the main hospital to cut FS’s, which is 2 blokes away. One of the tech’s was gone for 3 hrs because the pathologist performing the BM was extremely slow. That left 1 tech and myself to cover bench duties. We are currently getting our (+) charged slides from Cardinal Health and they have little or no charge. We currently have 2 Ventana IHC Instruments, as well a Special Stainer. We are having tissues falling partially and completely off even on the SS Stainer. Years ago when I worked at Biocare Medical I used to QC our Kling-On Slides before selling them by checking to see if the sections indeed stuck to the slides without any lifting on each lot. REMOVING STAINS FROM HANDS AND CLOTHING Some stains can be removed form the hands by using *Erado-Sol. Dilute Chlorox (or similar bleach) and Erado-Sol are useful for removing stains from white clothing. Care should be exercised in removing stains from colored clothing. To remove the following stains use the solutions indicated. 1.Alcian blue - to remove from the hands use the acid alcohol solution. For cleaning glassware try using dilute Chlorox or acid alcohol. If the stain resists, use them alternatively. 2.Auramine-rhodamine - acid alcohol. 3.Basic fuchsin - Erado-Sol. 4.Carmine - acid alcohol. 5.Chromic acid - concentrated aqueous sodium thiosulfate containing a few drops of sulfuric acid. 6.Crystal violet - Erado-Sol or Chlorox. 7.Eosin Y - saturated aqueous lithium carbonate. 8.Fast green and similar acid dyes - saturated aqueous lithium carbonate. 9.Hematoxylin (alum and iron) - Erado-Sol or acid alcohol. 10. Iodine - 5% sodium thiosulfate. 11. Methylene blue - acid alcohol, Erado-Sol or Chlorox. 12. Nuclear fast red - acid alcohol. 13. Picric acid - saturated aqueous lithium carbonate. 14. Potassium permanganate - 5% oxalic acid. 15. Schiffs solution - Erado-Sol or Chlorox. 16. Silver deposits - Lugols iodine followed by 5% sodium thiosulfate. 17. Sudan stains (Oil red O, Sudan black B) - acetone. 18. Thioflavin T - acid alcohol. *American Scientific Products ___ Histonet mailing list Histonet@lists.utsouthwestern.edu http://lists.utsouthwestern.edu/mailman/listinfo/histonet

Re: [Histonet] Supervising a histologist
We are CLIA/CMS certified only. My lab assistant only has a high school degree and never had any background in any sciences. She knows nothing about trouble-shooting, nor theory and practice. She has worked primarily in clerical roles for our GI practice for 13 years. In the past 1.5 years, I trained her to accession, process, embed, cut, stain, and coverslip. We do not do IHC here. Our CEO/GI, MD said she could oversee our new HTL during my absence. Re: [Histonet] CAP question ANP.12500
We keep ours for one week after final report. There have been past issues as to tissues submitted from our endoscopy center not matching up to our gross exams. Gastric tissue included with esophageal tissue, I account for every MINUTE fragment, so there were absolutely floaters from somewhere!. We account for everything when I gross the tissue! I have a embedding log sheet which accounts for every piece submitted, as well as who embeds it, cuts, it, etc.. as well as any comments which just don’t match up to the gross. [Histonet] RIP Wanda Platt Jones
My feelings for Wanda Platt Jones: Although I am Buddhist, and believe in reincarnation, I am untuned to all religions! In Wanda's case, perhaps, she was needed to be with her GOD to help serve in the best way possible for the ultimate good of everyone. She had an energy and presence which was focused unconditionally for true GOOD and LOVE for everyone. Wanda to me is a True Angel; who now is looking down on us, and helping us to become better human beings. Anyone she came in contact with benefitted from her remarkable love, sweetness, and acceptance. I will truly miss her, but I know she has not left us. She is hugging and guiding us, and showering us with her love and sweetness. She is sitting in a Prestigious Histology Chair in the Sky with all the Greats, such as the likes of Chuck Churukian. He was another Great Christian Histologist, and a Very, Dear Friend! XoXo ___ Histonet mailing list Histonet@lists.utsouthwestern.edu http://lists.utsouthwestern.edu/mailman/listinfo/histonet

Re: [Histonet] Marciafava-Bignami Disease I have re-logged into the system and maybe this will now communicate with Histologists out there, and hope that I do not get twice as many emails from Histone
GP’s are fine for general health issues, but I would certainly get more conclusive tests done by a neurologist, as well as contacting the best worker’s comp attorney in your area who has dealt with chemical exposure cases. Studies show we are #1 in this country for the most hazardous professions. It's safe compared to when I 1st started in this field in 1965! Well, I am a dinosaur from back in the day when we had inadequate ventilation, a shortage of fume hoods, inadequate education on hazards, safety and PPE's. In 1979 I started to work at OHSU in the surgical path lab, as well as doing research projects. We made up all of our own HE's and special stains from scratch, as well as made up our own 10% NBF in 55 gallon drums without fume hoods, ventilation, masks or gloves. I also worked with Glyco-Methacrylate embedded tissues without hoods or gloves! Since it was a medical school, we did every special stain under the sun and dealt with about every chemical, reagent, acid and stain you could think of! We also smoked cigarettes and drank coffee in the lab while we embedded and cut! We sure were a naive group back then! In the early days, the facilities I worked in never had MSDS information available. In 1989, while at Emanuel Hospital, Portland, OR, I researched and compiled the MSDS information on all the chemicals, reagents, acids and stains that we used. That was the 1st eye opener to what me and my fellow histologists dealt with on a daily basis. In 1988 I had base line tests done because I was having issues with dizziness, balance, reflexes, and short term memory loss. In 1992, I had extensive neurological tests done, as well as a sural nerve bx taken from my right ankle. It was found that I had nerve damage, loss of balance, no reflexes, numbness in my fingers, hyper sensitivity and reduced feeling on my right side, hearing loss in my left ear, and an aedes pupil in my right eye. It was concluded these were the results from exposure to multiple toxic chemicals in an extremely small room with excessive heat and NO VENTILATION at current hospital I was working at. These health issues are irreversible. I just deal with it. I was the 1st person who won a case for this in the state of Oregon, but it had consequences. I won the battle, but lost the war! I now have Primary Biliary Cirrhosis (PBC). I was 1st diagnosed in 2008 for PBC. I think this condition was caused by my continual exposure to Multiple Toxic Chemicals. You may, or may not agree. A huge amount of the chemicals we deal with in the histology lab targets the liver and is absorbed through the skin or is inhaled. Here is the link for PBC. http://www.liverfoundation.org/abouttheliver/info/pbc/ http://www.liverfoundation.org/abouttheliver/info/pbc/ Akemi Allison BS, HT/HTL (ASCP) Pathology Manager Monterey Bay GI Consultants Laboratory 23 Upper Ragsdale Drive, Suite 200 Monterey, CA 93940 W: Email: aalli...@montereygi.com mailto:aalli...@montereygi.com On Aug 4, 2015, at 4:47 AM, Edmondson David (RBV) NHS Christie Tr via Histonet histonet@lists.utsouthwestern.edu wrote: __ From: Edmondson David (RBV) NHS Christie Tr Sent: 02 August 2015 16:40 To: 'histonet@lists.utsouthwestern.edu' Subject:Marciafava-Bignami Disease Hello out there, I have been working in Histology for forty years and am wondering at a cause for this disease. Addition to work has been my only problem. They have tested a variety of bodily functions, touching my nose and foot movements together with assessing my ability to ride my motorcycle and make toast and tea, and then they let me off the acute neuro ward. My bike was chained up outside and making my way between lines of static traffic should have been a good sign. But I wonder if it was his exposure to jet fuel, etc. as he was a jet mechanic for 20 years in the USAF plus another 15-20 with a private company. So far my liver has been stable even getting a bit better to the point of being just a hair above the normal range. I do everything under the sun in my lab. Mohs, Grossing, plus all the routine stuff of cutting and staining, etc. I have been able so far to keep the repetitive injuries at bay, and Dansco shoes have saved my feet from the Plantar Faciitis (had that about a year straight at one point.). We had managed to keep xylene out of our lab for years by using Propar, but one of the Pathologists insisted on having a tape coverslipper. We still use it as little as possible. I had a co-worker, when she was pregnant wasn't allowed to gross (hence contact with formalin). Because of the NASH I get blood tests done every 6 months. It will be interesting to see if they have changed possibly due to the exposure to xylene, which I haven't had exposure to for years otherwise. I have worked in the field for over 20 yrs, and was pregnant while working in a small lab. Mostly everything was manual, and I did not use all of the safety precautions I should have (my fault). My son had severe speech and language delay as well as a language processing issue when he was smaller, and now stutters. I have often wondered (as most parents would) if my lack of precautions may have contributed. We stress the safety issues to the students daily, to get it all in their heads. I often tell people that I am so happy because I work with xylene, and I feel real good. So far, I don't see any noticeable symptoms of anything wrong, just the usual-decreased olfactory sensing, repetitive motion issues, plantar fasciitis, and varicose veins. Hair loss was an issue at one time, but that was attributed to PCOS. Now that I am not in the lab full time anymore it has decreased since I went natural (I'm African-american). One thing I have noticed is my C-reactive protein is high, and I take a statin for that. I have no liver issues, and my functions appear to be normal. Next time I go in for a physical, I will have my workplace hazards documented in my EMR. I will look up Primary Biliary Cirrhosis (PBC), to get some more information, especially since I was born jaundiced and I am anemic. Sincerely, Toysha N. Re: [Histonet] GI Pathology Req's
Just an FYI- We do not have a LIS System! The GI CEO and administrator DOES NOT want to invest in a LIS System! Our pathologists are contracted, so they just go with the flow… which is difficult for me to have support to upgrade our system. As usual, we are an after thought! When I came here 4 years ago, I thought I was in a time machine and had been transported back to 1989 before they had lab computers or LIS Systems! We are currently using “PCM”. It is geared towards GI MD’s and their practice. It is a system which is used for scheduling and all patient information. It interfaces with the billing system. This system was in place before I got here. Currently, my assistant has to create a Path Report and type in gross, etc… then the pathologist types in his microscopic findings and sign’s the report. Everything is hard copy….. I have basically time warped to before the 90’s when I created all these workload recording forms up. We have hard copy req forms which are hand written by the nurses in endoscopy with all the patient and specimen information. We do have an automated cassette printer, but we hand write slides. We hand write an embedding log with # of number of pieces in the cassette, etc…for QC purposes. I hand write CPT charges on a hard copy and submit to the billing dept. Nothing is bar coded, etc…. It is better now that I have an assistant. The endoscopy center has upgraded to “Provation”. It interfaces with our current “PCM” (Prime Clinical Management System) which is antiquated! The new computer system for scheduling and billing is Athena Healthcare” it is geared for billing and appointment scheduling etc We are also interfaced with Elkay? and are in training now and go live August 11th! We are still going to have to do the same procedure with pathology as usual, but we have to create the templates again! They are not getting us a LIS system to interface! Oh wait, I’ve been a histologist for 50 years and still going strong! It’s frustrating, but at least I live in a beautiful area! [Histonet] P26 billing for IHC Professional component
Good morning Histoland: Happy Friday! I was asked by a friend to post this on histonet. They have a question for those of you who may be familiar with billing for the IHC professional component only. Here is the scenario; a small lab who does not perform IHC in-house sends a block to be cut and stained for IHC to a reference lab who performs a stain only IHC. The reference lab then sends the stained slides back to the ordering facility and their pathologist makes the interpretation of the stained slides. Arrangements have been made for the reference lab to charge the patients insurance company and/or patient for the technical component of the IHC stain. How would the requesting facility billing department charge for the pathologists professional component? Do they charge with a P26 CPT code only? Thank you in advance for your responses! [Histonet] Levels for Gastic and Esophogeal Bx's
Good morning Histoland: Happy Friday! I believe this question was asked last year, but I would like to revisit this again. I have worked in a variety of laboratories in my career, both small and large. Most of my experience was in Oregon with large hospital laboratories and University institutions. Back then, we cut (6) levels on all our bx’s, and picked-up (2) sections on each level. My current facility is cutting (3) levels and picking up (1) section on each level. I would like to ask what everyone else is doing. The reason I am asking is because our pathology director is requesting a change in our protocol to cut only (2) levels and (1) section at each level. [Histonet] CLIA Compliance Regulations for histology staff coverage
Good morning to all in Histoland! Hopefully, one of you can help me with a question which was proposed to me by a fellow histologist working in a very small GI histology lab. Here is what information she gave me: She is training a histology assistant in house to do all the histology technical responsibilities at the request of the GI doctors. She said her OJT assistant is doing a great job technically, but she has not yet registered in school to finish her AA Degree so she can sit for the HT exam. She has been working under her instruction for 1 year. Her lab is California State and CLIA licensed. The GI doctors want the assistant to cover for her while she is on vacation. The pathologist who is the medical director does not think that CLIA Regulations allows this and wants the specimens sent out during her absence. She needs the CLIA Regulations stating what the guidelines are so her lab is complying to regulations. She didn’t want to call CLIA because it would send a RED FLAG up. I also didn’t want to call them because they may ask me what the name of the lab was that was proposing this question. I told her that this was most likely illegal, but she needs it in black and white. Do any of you have that information or a web link to go to at your finger tips? She needs the regulations by next week. Tony: I actually was going to suggest this. You beat me to it! I used to do GMA on 1.5 microns sections and did a Harris hematoxylin and eosin/phloxine routinely (by the way, this was not recommended back in 1980. Gills hematoxylin was the norm!). To get the results I wanted, I would routinely oxidize the slides in 1% periodic acid and rince in running tap water before staining in hematoxylin. the staining was intensified and was beautiful! I came up with this because my PAS's with hematoxylin counterstains looked better than routine HE's. I figuired it was worth doing the GMA's routinely using 1% periodic acid since we didn't do that many. Funny how we stumble on our modifications! Science never sleeps!! Re: [Histonet] Re: HE staining question
Oh by the way, the reason I did a Harris hematoxylin and eosin/phloxine on GMA (plastics) was because our pathologists at OHSU hated Gills and were used to seeing Harris hematoxylin on all of our regular surgical and autopsy slides. It is recommended to use DI water for your water bath, especially when cutting tissues for IHC. Tap water has all kinds of bacteria, as well as potential chorine, rust, lead and other components which might have adverse affects to your histologic testing. If you have an opportunity i highly recommend attending As a good housekeeping practice, I always recommend cleaning your water baths with a soapy detergent after use, rinsing with tap water, drying residual water from the water bath and either turning upside down, or covering to prevent dust particles. Re: [Histonet] Histotechnician Salary 2
Tim: I have worked in California since 1998, both in the SF Bay area and LA. The cost of living is extremely high in these two areas compared to the rest of California, and certainly higher than most of the country. Working as a manager in clinical and bio-tech settings, I found it difficult to find a histologist that was well rounded in all aspects of histology and IHC labs. The histologists that were suitable to hit the ground running were compensated accordingly. The average pay range paid for a registered HT/HTL was $25.00 to $40.00 per hour. There are a few bio-tech companies in the SF Bay area that hire unregistered histo techs and pay them $15.00 an hour. They primarily cut control tissues and do the less complicated assignments, I also know of several clinical histology laboratories in LA and SF that hire unregistered histo tech's and pay them $15.00 to $20.00 an hour. You have to understand that we do not require registered histo techs to work in a lab as long as they have a registered supervisor. The quality of work is compromised because these techs do not have the knowledge of theory and practice, You pretty much get what you pay for. I was told a couple of years ago from a very good source that a recent graduate of a LA histology program who passed their HT registry with a year of experience was paid $25.00 an hour to start. Re: [Histonet] Cooling paraffin blocks with ice VS. Freezing Spray
Well said Rene! In my 40 plus years as a supervisor, manager, technical support manager, and consultant; I have come across a broad spectrum of experienced and inexperienced histologists. Start looking for another place although do not expect that your ideas will always be well received. Older, trained on the job, and with lots of experiences supervisors usually are not very open to suggestions, especially when those ideas conflict with what they are used to do because they do not know the scientific basis of what they are doing. The less open to suggestions a person is, the more ignorant they are likely to be. René J. From: Jenny Vega histotech...@gmail.com To: joelle weaver joellewea...@hotmail.com; histonet@lists.utsouthwestern.edu Sent: Saturday, September 29, 2012 6:26 PM Subject: Re: [Histonet] Cooling paraffin blocks with ice VS. Freezing Spray Thanks everybody for your answers. I cant respond them all but I concluded that the best way to get good sections is too chill the blocks on ice because I agree that it facilitates the process. I really don't understand why my supervisor depends so much on freezing sprays to cut and the pathologist has never complained about artifacts caused by them but I do believe that they are present because I have seen them getting formed. It makes sectioning difficult because you try to get sections free of holes and that contributes to the problem. At my lab is the same thing. My supervisor is in charge of the embedding and she just use the ice only for hardening the paraffin block. We don't have a standard embedding center with cold plate. Since is a small lab we just have a heating plate where we handle the specimens and place them in the molds and we cool them on ice trays. After they are removed from the molds they are placed on the counter in numerical order and they reach room temperature and get warm. I do think that if they get cold and moistened since the beginning it can facilitate the sectioning process except for certain tissues that are not well processed. In my lab I change the reagents in the tissue processor weekly but they get dirty too quickly because we processes a lot of breast and colon tissue so is hard to get perfectly processed tissue daily. We are under a tight budget and we can't waste materials too quickly. This situation with my supervisor has caused me a lot of frustration. I have noticed that every tech has their own method to do things but unfortunately there are people who are not receptive to new ideas and they immediately criticize and say you are wrong specially if you are young and you have recently started your career. I have though on several occasions that all I learned in histotech school have been worthless because everybody in the lab does things differently and this has made me question my ability of being a good tech because I have experienced difficulty using their microtomy technique but I have realized that I am not wrong. Another issue was the use of microtome blades. Since I started my supervisor has told me to use the minimum amount of blades as possible because we are under a tight budget but I have noticed that some of those blades are of poor quality and since we cut a lot of tissues that are hard, calcified or have sutures they wear the blades too quickly. It's hard to cut many blocks using only one blade that you use to trim and section. I have realized that I am successful obtaining good sections when I chill the blocks on ice for a couple of minutes and change the blade immediately after encountering difficulties with a block. It's not worth to sacrifice the quality of the samples just to be cheap and save some money. My supervisor is a great person and tech with a lot of experience but she is not very open to new ideas. Her demands to save money can be unrealistic when the correct technique is not being used . With her technique of course you are going to waste excess of freezing spray and blades but she doesn't understand this. Perhaps I need a change of environment which is unfortunate but is difficult to work like this. Thanks everyone

[Histonet] Re: Standardization??? 3, 000 Breast Cancer Specimens from Quebec Breast Cancer being retested
From what I have experienced in my observations of various labs; numerous labs do not follow proper standardization practices which are set by FDA guidelines for Her2-neu. Prognostic markers are particularly susceptible to heat. Baking slides in excess temperatures (over 62 degrees C) for prolonged lengths of time can also drop the antigenicity of the test tissues a full grade. There has been documentation of this through publications. Temperatures MUST be monotored and recorded. The Antigen Retrieval Process is very important and QC measures MUST be followed. Internal temperature and pressure MUST be met and recorded. QC monitor strips will record and show any irregularities in this step. If the pressure and temp has not been met, the test should be repeated. A lot of IHC techs do not monitor this process. Unfortunately, internal laboratory standardization can vary from shift to shift. Just think about non- standardization practices throughout the country because of under- education and strict adherence to established guidelines. Hello Histoland! Does anyone know how to get in touch with Rita Humphrey, previously with Childrens Hospital, Birmingham, AL? I lost touch with her after she left Childrens and would like to re-connect. [Histonet] GREAT B5 substitute
Hi All, I used to use B5, Zenker's and Helly's for years on LN and BM biopsies. I wanted to stop using mercury in our department, but didn't want to compromise the quality of the results we were getting. I was interested in using fixatives and decalcification products that would not effect IHC results. At the suggestion of Dr. Elaine Jaffe, Chief Hematopathologist from NIH, I tried 2 products from BBC Biochemical. I did parallel studies on bilateral BM bx's using B•Plus Fixative™ and RapidCal•Immuno™, and then B5 fixative and EDTA decal solution, and then Zenker's fixative. We then did parallel studies for IHC. We ran a panel of Ab's on specimens which were treated with each of the solutions. The pathologists concluded that B•Plus Fixative™ produces superb fixation for hematopoietic tissues and the RapidCal•Immuno™ does not interfere with IHC staining. In some cases, the results on the IHC's were better on some of the Ab's using these products too. It only takes 2-3 hours of fixation time for BM biopsies. After prefixing, they can go directly into RapidCal•Immuno™ for 45 minutes to 1 hour, washed in running water for 5 minutes, and then continue to be processed as normal starting off in 10% formalin. Just a note: This product is a rapid decalcifier designed to preserve immunogenicity at a maximum while accomplishing rapid decalcification. It is designed specifically for bone marrow biopsies and LN biopsies. By the way, I am not on BBC's payroll. I am always on a quest to improve patient care! Re: [Histonet] bone marrow biopsy decals
Hi Diana, I recently made a histonet posting Great B5 substitute which also addresses BM decal. Since there are a number of companies who sell decal solutions with similar names, I am not sure you tried the products from BBC Chemical. It is best to use both products together. My post goes into my findings. Also we a decal that will minimize the leeching out of iron. If any out there has any suggest, please let me know. Thanks in advance!
