I all reality none is good. B= oth alkanes or D-Limonene are either
   as  dangerous  or more than xylene, and = all perform below the xylene
   standard.
   
Any  one  requires  modifications=  of  all procedures, they seldom
   dewax  adequately and all constitute a proce= ssing compromise. Many
   cannot de recycled.

The optimal solu= tion to eliminate xylene consists on:
   
1- dehydrate with 2-propanol (o= r pure isopropyl alcohol or IPA)
   
2-  do  not  use  any  ante-medium  and  =  go from IPA directly to
   paraffin wax or ideally to mineral oil:IPA at = 1:1
   
3- infiltrate as usual
   
4-  dewax  sections with 2%aq. sol. of= dishwater soap at 90ºC
   for 1 min twice
   
5- stain as usual
   
=  6-  dehydrate stained sections in a convection oven at 60ºC
   for 5 minut= es
   
7- coverslip as usual.
   
As  you  can  see  xylene is nowhere to b= e found in this sequence
   that produce optimal quality slides.

Good  Morning, We have two Leica HE stainer( old xt-5010 an= d new
   5020). We do use : Gill's 3 for 6 minutes,
   
4% Glacial ACETIC Ac= id 40 seconds
   
Bluing  :  45  sec.  We done this for years!! last few mont= hs our
   Pantologists  are complaining about GI Biopsies that they have TOO mu   ch 
blue in cytoplasma and the Mucin is too purple and blue? Please any
   sugg= estion??
   
Mean  while  we  are trying the NEW kit from Leica for HE = , they
   think  that is TOO pale?any of you has tried this new stain from Leci   a?

I would first check with y= our alcohol and xylene suppliers to see
   if  they  changed anything.? When som= ething?goes south it is usually
   because the manufacturer have changed.? Thi= s happened with my silver
   stains.

You  may  have to go back to = the basics- harris hematoxylin, acid
   alcohol,  and  eosin y.? Richard Allen 7= 211 hematoxylin and the rest
   of their staining is excellent in these situat= ions until you can get
   it back under control.? If you send them slides they= will work a time
   out for you.

If  you are using Leica's selec= tech, which is what I started this
   lab  out  with because I was heavily sampl= ed out,?it has blue nuclei
   instead  of  the traditional purple.? I do 5 minut= es in hematoxylin,
   30  dips  in  clarifier  and 1 minute in blue, 55 sec in eos= in but I
   have  (2)  95s  after.? They also have a tech dept to help.? I have a   
Leica autostainer too.

[Histonet] used lab equipment

   I posted last week and got several responses. Anyone interested i= n
   obtaining  this  equipment,  please  email  me what you are willing to
   pay=  BRfor  it.  Whoever  buys  must  also  agree  to  pay  for all
   packaging,
   handli=  ng,  freight  and  shipping costs related to the equipment. I
   will
   accept e= mails related to this equipment until Friday 11/9/12

[Histonet] Disposal of used lab equipment

   We have upgraded some of our equipment in recent months and repla= ced
   some VERY OLD equipment that is just taking up space now.

   I have contacted some of the used equipment man= ufacturers and no one
   is interested in taking this equipment off our hands.
   




   


   My  question  is  how do you dispose = of used histology equipment.  I
   don't  know  if our local landfill will = take it.  Is there a service
   out there who will come haul it away?
