I have a small domain with 3 Windows 2000 DCs and 2 Exchange 2000 mail

During the day? It depends what other services are running on the DCs. As long
as the switch is smooth (enable new first, disable older second) nobody will
notice it

I've done it during the day at a customer, but without much experience I
would strongly recommend doing it after hours! There are always minor things
which might happen, and without experience you don't know how to respond to
them right away, so give yourself the rest and peace of after hours.
If the Exchange server is on a domain controller, you'll have to upgrade
Exchange first.
